高效液相色谱法同时测定蒙药复方协日嘎-4中4种成分的含量 被引量:3

摘要 目的建立HPLC法同时测定蒙药协日嘎-4中4种成分的含量。方法采用Elite C18色谱柱(250mm×4.6 mm,5μm),流动相为乙腈(A)和0.5%醋酸水溶液(B),梯度洗脱,流速为0.8 m L·min-1,柱温30℃,检测波长270 nm和420 nm,进样量为10μL。结果黄芩苷、双去甲氧基姜黄素、去甲氧基姜黄素、姜黄素的质量浓度在线性范围与其峰面积有良好的线性关系(R>0.9998);平均回收率分别为97.2%、97.1%、96.4%、96.8%,RSD分别为1.1%、1.4%、0.86%、1.0%;平均含量分别为0.0080%、0.0377%、0.0292%、0.0631%。结论经方法学验证,本方法可用于协日嘎-4中4种化学成分含量测定。 Objective To establish an HPLC method for the simultaneous determination of 4 effective components in Mongolian medicine compound Xieriga-4. Methods Elite C18 column (250 mm×4.6 mm, 5 μm) was used with a mobile phase of acetonitrile (A) and 0.5% acetic acid aqueous solution (B) with gradient elution at the flow rate of 0.8 mL · min - 1. The column temperature was 30 ℃ , the detection wavelengths were 270 nm and 420 nm, and the injection volume was 10 μL. Results Mass concentration and the peak area of baicalin, bis-demethoxy curcumin, demethoxy curcumin and curcumin showed a good linear relationship with correlation coefficient 〉 0.9998. The average recovery was 97.2%, 97.1%, 96.4% and 96.8%, with RSD of 1.1%, 1.4%, 0.86% and 1.0%, and the average content was 0.0080%, 0.0377%, 0.0292% and 0.0631%, respectively. Conclusion Methodology validation shows that the established method can be used to determine chemical constituents in Mongolian medicine Xieriga-4.
