加味黄芪赤风汤对阿霉素诱导肾小球足细胞凋亡的影响 被引量:8

摘要 目的:探讨加味黄芪赤风汤对阿霉素诱导的肾小球足细胞凋亡的影响。方法:体外培养小鼠肾小球足细胞,以阿霉素(adriamycin,ADR)诱导制作足细胞损伤模型。按随机数字表法分为空白组、模型组、替米沙坦组及加味黄芪赤风汤高、中、低剂量组(简称中药高、中、低剂量组),采用CCK-8法检测各组足细胞存活率,流式细胞仪检测各组足细胞凋亡情况,并采用EXPO32v1.2软件分析足细胞凋亡率,透射电镜下观察各组足细胞凋亡超微结构。结果:与空白组比较模型组足细胞存活率明显下降,与模型组比较中药高、中剂量组足细胞存活率均明显上升,中药低剂量组无明显升高;与空白组比较模型组早期凋亡率明显升高,与模型组比较中药各组及替米沙坦组早期凋亡率均明显下降,中药高、中剂量组与替米沙坦组比较差异无统计学意义,与低剂量组比较差异有统计学意义;模型组较空白组足细胞明显皱缩、胞浆内细胞器减少,染色质边集、凝聚成块、核碎裂,形成凋亡小体,中药各组及替米沙坦组较模型组凋亡超微结构均有不同程度改善。结论:加味黄芪赤风汤对ADR诱导的足细胞损伤具有保护作用,并呈现一定的量效关系,其保护作用可能与该方能够抑制足细胞的过度凋亡有关。 Objective: To explore the effect of Modified Huangqi Chifeng Decoction( MHCD) on the expression of apoptosis in glomerular podocytes induced by Adriamycin. Methods: Using in vitro cultured podocytes,produceing type of podocyte injury induced by adriamycin( ADR) and intervening with MHCD. This research was divided into 6 groups( the blank group,the ADR group,the Telmisartan group,and the high,middle and low group of MHCD),CCK-8 was used to detect changes of podocyte viability in different groups of serum containing. Transmission electron microscope was used to detect apoptosis. Flow cytometry was used to calculate the rate of podocyte apoptosis. Results: Compared with the blank group,the podocyte viability of the ADR group was significantly decreased; compared with model group,the high and middle group of MHCD and the Telmisartan group significantly increased the viability of podocyte,However,the low group wasnot significantly increased; Apoptotic bodies: compared with blank group,the number of apoptotic bodies increased significantly,volume increased significantly,organelles decreased in ADR group; compared to ADR group,the quantity and volume of Apoptotic bodies reduced more obvious in other groups; compared with the blank group,the early apoptosis rate was significantly increased in ADR group; compared with the ADR group,the early apoptosis rate in the high,middle,low group of MHCD and the Telmisartan group was significantly decreased; compared with the high and middle group,There was no statistical difference in the Telmisartan group,however,the early apoptosis rate was significantly decreased in low group of MHCD group. Conclusion: MHCD can protect the function and structure of the podocyte by inhibiting excessive apoptosis of podocyte probably.
