响应面优化酶协同超声提取四角菱角壳多酚工艺 被引量:1

Optimization of Enzymatic Ultrasonic-Assisted Extraction of Polyphenols from Pericarps of Trapa natans L. Var. incisa Makino by Response Surface Methodology

摘要 目的:优化四角菱角壳中多酚的酶协同超声提取工艺。方法:采用酶协同超声提取法,在单因素试验基础上,以多酚提取量为应变量,以纤维素酶浓度、超声提取时间、提取温度为自变量,采用响应面设计方法优化四角菱角壳中多酚的提取工艺。结果:最佳提取工艺为用pH 4.5的1.86%纤维素酶溶液30 m L·g-1预处理30 min后,在53℃下超声提取26 min,在此条件下多酚提取量为161.3 mg·g-1,与模型预测值(158.14 mg·g-1)相符,相对标准偏差2.00%。与乙醇超声提取和回流提取相比,酶协同超声提取多酚提取量显著提高。结论:响应面法优化四角菱角壳多酚的酶协同超声提取工艺方法简单、提取效率高、可预测性良好,值得推广使用。 Objective: To optimize theenzymatic ultrasonic-assisted extraction of polyphenols from the pericarps of Trapa natans L. Var. incisa Makino. Methods: Based on the single factor results,the independent variables such as cellulase concentration,ultrasonic time and extraction temperature were optimized by response surface methodology(RSM) with polyphenols yields as dependent variable. Results: The optimal conditions obtained by RSM with enzymatic ultrasonic-assisted extraction were as follows:water-material ratio of 30 m L·g-1,pH = 4. 5,1. 86% cellulase pretreated 30 min,extraction temperature 53 ℃,and extraction time 26 min. Under this optimal conditions,the experimental polyphenols yields were161. 3 mg·g-1,which was well agreed with model predictions of 158. 14 mg·g-1with RSD 2. 00%. Conclusion: The extraction process of polyphenols from Trapa natans L.Var. incisa Makino by enzymatic ultrasound-assisted was stable and efficient with high precision and good predictability.
