胃幽康胃漂浮片抗幽门螺杆菌和抗胃溃疡的实验研究

Experimental study on antimicrobial activities against Helicobacter Pylori in vivo and in vitro and antigastric ulcer of the Weiyoukang intragastric floating sustained-release tablets

目的:研究胃幽康胃漂浮片对幽门螺杆菌和实验性胃溃疡的影响。方法:采用体外抗幽门螺旋杆菌试验、大鼠幽门结扎胃溃疡抑制试验、大鼠吲哚美辛胃溃疡抑制试验、大鼠棉球肉芽肿试验、小鼠耳二甲苯致炎试验和小鼠醋酸扭体镇痛试验,对胃幽康胃漂浮片进行了初步的药效学评价。结果:胃幽康胃漂浮片在体外对HP的生长具有显著抑制作用,其MIC值为18.75mg·mL^(-1);胃幽康和阳性对照药阿莫西林组均能有效清除HP感染小鼠模型胃内的HP。胃幽康胃漂浮片能明显抑制幽门结扎致大鼠实验性胃溃疡和吲哚美辛致大鼠胃溃疡,且有一定的量效关系。胃幽康高中低3个剂量组均能不同程度的降低醋酸引起的小鼠疼痛反应,而且能明显抑制棉球引起的大鼠慢性肉芽肿增生和二甲苯引起的小鼠炎症反应。结论:胃幽康胃漂浮片具有杀灭幽门螺旋杆菌、抗胃溃疡、抗炎和镇痛的药理作用。

OBJECTIVE To study antimicrobial activities against Helicobacter Pylori in vivo and in vitro and anti gastric ulcer of the Weiyoukang intragastric floating sustained-release tablets（WYK-IFSRT）.METHODS The minimum inhibitory concentrations（MIC）of WYK-IFSRT and amoxicillin were determined by using the broth dilution method.The mice HP infection model had been established successfully to study inhibition action of WYK-IFSRT against the growth of HPin vivo.Antigastric ulcer action of WYK-IFSRT was studied by pyloric ligation gastric ulcer in rats and indomethacin in rats with gastric ulcer.RESULTS The MIC of WYK-IFSRT was 18.75 mg·mL^-1.WYK-IFSRT inhibited the growth of HP in vivo.WYK-IFSRT reduced ulcer index of pyloric ligation gastric ulcer in rats and indomethacin in rats with gastric ulcer,WYK-IFSRT relieved the mouse inflammation induced by xylene（P〈0.01）,decreased rat granuloma weight induced by tampon（P〈0.01）,and lessened the swiveling induced by glacial acetic acid（P〈0.01）.CONCLUSION WYK-IFSRT is effective against the growth of HP in vivo and in vitro,and gastric ulcer,and has anti-inflammatory and analgesic actions.
