牛至油注射亚微乳中3种成分的含量测定

Contents of three compounds in oregano oil submicron emulsion for injection

目的:通过HPLC法测定牛至油注射亚微乳中3种成分(麝香草酚、香荆芥酚、对聚伞花素)的含量。方法:采用Kromasil 100-5C18column为分析柱,流动相为乙腈-0.2%磷酸水溶液(54∶46),检测波长274nm,柱温30℃,流速1.0mL·min^(-1)。结果:香荆芥酚在0.48~7.2μg的范围内线性关系良好,线性方程为:Y=319.09 X+0.425 1,r^2=1;麝香草酚在1.04~15.6μg的范围内线性关系良好,线性方程为:Y=690.09 X+0.353 4,r^2=1;对聚伞花素在1.88~28.2μg的范围内线性关系良好,线性方程为:Y=115.15 X^(-1)8.037,r^2=0.999 9。测得牛至油注射亚微乳中香荆芥酚的含量为1.033mg·mL^(-1),麝香草酚的含量为4.561mg·mL^(-1),对-聚伞花素的含量为0.185mg·mL^(-1)。结论:该方法快速、准确、重复性好、分离度高,可同时测定3种成分的含量,可用于牛至油注射亚微乳的质量控制。

OBJECTIVE To determine the contents of three compounds（carvacrol,thymol,P-cymene）in oregano oil submicron emulsion for injection by HPLC.METHODS Kromasil 100-5 C18 column was used as the analytical column,with the mobile phase of acetonitrile-0.2% phosphoric acid solution（54∶46）,the detection wavelength at 274 nm,the column temperature at 30 ℃,the flow rate at 1.0 mL·min^-1.RESULTS In range of 0.48-7.2μg,carvacrol had a good relationship,linear equation：Y=319.09 X＋0.425 1,r^2=1;in range of 1.04-15.6μg,thymol had a good relationship,linear equation：Y=690.09 X＋0.353 4,r^2=1;in range of 1.88-28.2μg,P-cymene had a good relationship,linear equation：Y =115.15X-18.037,r^2=0.999 9.The content was 1.033 mg·mL^-1 for carvacrol in oregano oil submicron emulsion for injection,4.561 mg·mL^-1 for thymol,0.185 mg·mL^-1 for P-cymene.CONCLUSION The method is rapid,accurate and reproducible,with a high resolution,can simultaneously determine the contents of three components.It can be used for the quality control of oregano oil submicron emulsion for injection.
