摘要
目的通过加味小柴胡汤抗菌抗炎有效物质部位的体内过程有效成分研究,进一步诠释该中药复方的药效物质基础。方法将前期研究确定的加味小柴胡汤抗菌、抗炎的两个主要有效物质部位,即酚酸物质部位和苷类物质部位,分别灌胃给药大鼠,不同时间段取血,制备含药血清供试品。采用LC-MS现代分析方法,分离和鉴定各有效物质部位的入血成分,获得血清中移行成分的信息。结果从酚酸物质部位给药血清中分离鉴定了8种化合物,其中4种为原型成分,其它4种为代谢产物;从苷类物质部位给药血清中分离鉴定了12种化合物,其中6种为原型成分,其它6种为代谢产物。结论文献报道表明,从酚酸物质部位和苷类物质部位的给药血清中所鉴定的化合物均具有抗菌抗炎作用,从而进一步诠释了加味小柴胡汤的药效物质基础。
Objective To study the constituents in process in vivo of antibacterial and anti-inflammatory active fractions of modified Xiaochaihu decoction to further illustrate the material foundation of medicinal effectiveness of the decoction. Methods The two active fractions of antibacterial and anti-inflammatory of modified Xiaochaihu decoction were respectively given to rats. Blood samples were taken at various times to prepare serum samples. The aim was to separate and identify the components of ingested into blood and obtain the information of blood transitional components with LC-MS technique. Results 8 compounds were isolated from the serum of phenolic acid sites, of which 4 were prototypes, the other 4 were metabolites. 12 compounds were isolated from the serum of glycosides, 6 of them were prototypical components, and the other 6 were metabolites. Conclusion Literature reports showed that the compounds identified from the serum of phenolic acids and glycosides had antibacterial and anti-inflammatory effects, which further explained the effective material basis of modified Xiaochaihu decoction.
