脾胃虚实证大鼠局部与整体Th平衡的研究

Study on Local and Integral Th Balance in Rats with Deficiency and Excess of Spleen and Stomach Syndrome

目的探讨脾胃虚实证候大鼠模型胸腺、脾脏、外周血Th1/Th2平衡的相关性。方法将60只SPF级SD大鼠随机分为3组,其中一组为脾虚组,一组为湿热组,一组为正常组,每组20只,雌雄各半,采用酶联免疫吸附法(ELISA)技术对大鼠模型血浆、胸腺、脾脏组织Th1/Th2细胞因子水平进行检测。结果在胸腺方面,与正常组对比,脾虚组的IFN-γ表达下降,IL-4表达和IFN-γ/IL-4比值升高,但均无统计学意义(P>0.05);湿热组的IFN-γ表达和IFN-γ/IL-4比值明显升高(P<0.05),IL-4表达下降,但无统计学意义(P>0.05)。在脾脏方面,与正常组对比,脾虚组的IFN-γ表达下降,但无统计学意义(P>0.05),IL-4表达明显下降(P<0.05),IFN-γ/IL-4比值明显升高(P<0.05);湿热组的IFN-γ表达升高,但无统计学意义(P>0.05),IL-4表达明显下降(P<0.05),IFN-γ/IL-4比值明显上升(P<0.05)。在外周血方面,与正常组对比,脾虚组的IFN-γ表达明显下降(P<0.05),IL-4表达升高,IFN-γ/IL-4比值下降,但均无统计学意义(P>0.05);湿热组的IFN-γ表达与IFN-γ/IL-4比值明显升高(P<0.05),IL-4表达下降,但无统计学意义(P>0.05)。结论脾虚组和湿热组在脾脏的IL-4表达均表现出下降趋势,IFN-γ/IL-4比值呈升高趋势,脾脏的Th1/Th2平衡在不同证型中可能均向Th1漂移。胸腺、脾脏、外周血等不同组织的IFN-γ/IL-4比值在脾胃虚实证候的呈现不同,提示不同部位的细胞因子表达差异性与不同证候的相关性,这可能是脾胃虚实证候的发病机制之一。

Objective To study the correlation between the Th1/Th2 balance in thymus,spleen and the peripheral blood of rats with deficiency and excess of spleen and stomach syndrome. Methods： Sixty SD rats were randomly allocated into normal group（NG）,Piasthenic syndrome group（PSG） and damp-heat syndrome group（DSG）. Every group had 20 rats,with ten males and ten females respectively. Thecytokine levels interferon（IFN）-γ,interleukin（IL）-4 in thymus and spleen the peripheral blood of rats were determined by enzyme-linked immune sorbent assay. Results： In thymus,comparing with NG,the expression of IFN-γ in PSG has decreased,while the expression of IL-4 and the ratio of IFN-γ/IL-4 in PSG have increased. But they didn＇t have statistic significance（P〈0. 05）. In DSG,the expression of IFN-γ and the ratio of IFN-γ/IL-4 have risen significantly（P〈0. 05）,while the expression of IL-4 has decreased with no statistic significance（P〈0. 05）. In spleen,comparing with NG,the expression of IFN-γ in PSG has decreased with no statistic significance（P〈0. 05）,while the expression of IL-4 in PSG has decreased significantly（P〈0. 05） and the ratio of IFN-γ/IL-4 in PSG has risen significantly（P〈0. 05）. In DSG,the expression of IFN-γ has increased with no statistic significance（P〈0. 05）,while the expression of IL-4 has decreased significantly（P〈0. 05） and the ratio of IFN-γ/IL-4 has risen significantly（P〈0. 05）. In the peripheral blood,comparing with NG,the expression of IFN-γ in PSG has decreased significantly（P〈0. 05）,while the expression of IL-4 in PSG has increased and the ratio of IFN-γ/IL-4 in PSG has decreased,nevertheless,with no statistic significance（P〈0. 05）. In DSG,the expression of IFN-γ and the ratio of IFN-γ/IL-4 have risen significantly（P〈0. 05）,while the expression of IL-4 has decreased with no statistic significance（P〈0. 05）. Conclusion： In Spleen,the expressions of IL-4 of deficiency and excess of spleen and stomach syndrome showed a decreasing tendency,and the ratio of IFN-γ/IL-4 shows an increasing trend. The balance of Th1/Th2 in spleen may have different syndromes. The expression of IFN-γ and IL-4 in different tissues such as the thymus,spleen and peripheral blood has difference between the deficiency and excess of spleen and stomach syndrome,which implies the correlation between the difference of cytokines expression in different organs and different syndromes. It may be one of the pathogenesis of deficiency and excess of spleen and stomach syndrome.
