益奥源胶囊降血糖药效学初探与分析

Preliminary Study and Analysis on Hypoglycemic Effect of Yi'aoyuan Capsule

目的:研究益奥源胶囊试验对小鼠降血糖的作用及胰腺病理组织学变化情况。方法:选取造模成功的2型糖尿病小鼠60只,随机分为模型对照组,阳性对照组,益奥源胶囊小、中、大剂量组以及喂普通饲料的正常对照组,每组10只。阳性对照组灌胃盐酸二甲双胍400 mg/kg,益奥源胶囊小、中、大剂量组分别灌胃益奥源胶囊混悬液3.0、6.0、12.0 g/kg,模型对照组灌胃和正常对照组灌胃等体积的蒸馏水,给药1次/d,连续给药40 d。在给药过程中每周定时对小鼠的体质量和血糖含量进行测量,末次测血糖后每组挑选2只接近血糖平均值的小鼠取胰腺作组织病理学观察。结果:与模型对照组比较,益奥源胶囊小剂量组的体质量在给药第1周和第3周后比较有所降低,第1周、第3周和第4周的血糖水平降低;胰岛病理组织学实验结果表明,益奥源胶囊能改善糖尿病模型小鼠胰岛细胞的功能,促进胰岛细胞的修复。结论:益奥源胶囊具有明显降血糖作用,并促进胰岛细胞的修复。

Objective： To study the hypoglycemic effect and pancreatic pathological changes of Yi＇aoyuan capsule on mice. Methods： The model of type II diabetes mouse were randomly divided into the model control group,the positive control group,the low,medium and high dose of Yi＇ aoyuan capsule group and the normal group with common feed,with 10 mice in each group. The positive control group was given 400 mg/kg metformin hydrochloride by gavage. Mice of Yiaoyuan capsule group were administrated by intragastrical gavage with 3 dosage of Yi＇ aoyuan capsule（ 3. 0,6. 0,12. 0 g/kg）. The model control group and the normal group received the same volume of distilled water intragastrically. The drugs were administrated once a day for 40 days. The weight and blood-sugar level of mice were calculated weekly. Islet tissue pathology and electron microscopy islet cell were observed after the last administration using two mice obtained from each group,of which blood-sugar level were close to the average. Results： Compared with the model control group,the weight of the first week and third week of the low dose group of Yi＇ aoyuan capsule increased（ 34. 15 ± 4. 21 g vs 38. 81 ± 4. 56 g,34. 70 ± 3. 78 g vs 39. 81 ± 5. 63 g）; blood-sugar level of the first,third and fourth week decreased（ 16. 47 ± 5. 43 mol/L vs 24. 59 ± 2. 70 mol/L,15. 60 ± 5. 73 mol/L vs 22. 42 ± 4. 96 mol/L,14. 37 ± 5. 25 mol/L vs 23. 38 ± 5. 18 mol/L）; The damaged islet cells of mice could be improved and the islet β cells could be repaired by Yi/aoyuan capsule. Conlusion： Yi/aoyuan capsule has a significant hypoglycemic activity and it also has a function to repair islet βcells.
