摘要
目的:基于血清药理学理论,探讨蒙药草果四味汤散对异丙肾上腺素(ISO)诱导H9c2心肌细胞损伤中的作用.方法:用不同浓度蒙药草果四味汤散含药血清(CG)预处理H9c2心肌细胞24h后,加入ISO.24h后MTT法检测细胞存活率;TUNEL染色法检测细胞凋亡率;Western blot法检测p38 MAPK及其磷酸化蛋白的表达.结果:CG预处理后显著提高ISO诱导的心肌细胞存活率,降低心肌细胞凋亡率.值得注意的是,CG预处理在ISO诱导的心肌细胞损伤模型中显著下调p38 MAPK蛋白的磷酸化水平.结论:CG对ISO诱导的H9c2心肌细胞损伤具有一定的保护作用,这可能与其抑制p38 MAPK磷酸化有关.
Objective: Based on the theory of serum pharmacology to explore the effect of Mongolian Amomum tsaoko four flavor soup(GC) against H9 c2 myocardial cells toxicity induced by isoprenaline(ISO). Methods: H9 c2 cells were pretreated with different concentrations of CG for 24 h then added ISO. Cell viability was detected by MTT assay.The rate of apoptosis was detected by TUNEL staining. The expressions of p38 MAPK and its phosphorylation were detected by Western blot. Results: Pretreatment with CG increased cell viability and decreased the cardiomyocyte apoptosis. It is worth noting that pretreatment with CG significantly down regulated the phosphorylation of p38 MAPK protein in ISO-induced cardiomyocyte injury. Conclusion: CG has protective effects on ISO-induced H9 c2 myocardial cells injury,the mechanism is likely to inhibiting the phosphorylation of p38 MAPK.
