心水通胶囊中药水提物的急性毒理学实验研究 被引量:3

摘要 目的:通过心水通胶囊中药水提物小鼠灌胃实验,研究该药物的毒理学反应。方法:分别以心水通胶囊中药水提物最大浓度和动物的最大胃容纳灌服给药,连续观察14 d,正常饮食组为对照组。观察小鼠毛色体征、活动情况、体重变化、尿量及尿液成分变化,检测血电解质、心肝肾功能以及部分内脏组织病理学变化。结果:心水通胶囊中药水提物的最大耐受量为204.24 g﹒kg^(-1),按60 kg人计算,相当于成人临床用量的近400倍。从第二天起,给药各组小鼠的尿量比对照组明显增多,且呈剂量依赖性;而外观行为,饮食体重、大小便和分泌物、血电解质、心肝肾功能与对照组比较无显著差异;给药各组心、肝、脾、肺、肾的病理切片未发现典型的炎症、增生或萎缩等病理学改变,形态与对照组相似。结论:心水通胶囊中药水提物对受试动物未出现无明显急性毒理学反应,可安全用于临床。 Objective: The acute toxicology was investigated after intragastric administration of the water extract of Xinshuitong Capsule to mice. Method: The difference concentrations of water extract according to the maximum stomach volume were intragastricly administered for continuous 14 days. The normal diet group was used as the control group. The alterations of the hair color, activity, weight, urine volume and its compositions were observed. The blood electrolytes change, the functions of heart, liver and kidney and some visceral histopathology changes were detected. Result: The maximum tolerance dosage of the water extract of Xinshuitong Capsule was 204.24 g· kg^-1, which was equivalent to nearly 400 times of adults (60 kg) clinical dosage. From the second day to the end of the experiments, the urine volume in the treated groups was significantly increased compared with that of the control group in a dose-dependent manner. However, the mice appearance, behavior, diet, weight, urine compositions, secretions, blood electrolytes, heart, liver and kidney functions showed no significance difference compared with the control group. No signs of typical inflammation, hyperplasia, shrink or other pathological changes in heart, liver, spleen, lung and kidney were found in each drug group. And the pathological condition was similar to the control group. Conclusion: The water b extract of Xinshuitong Capsule has no obvious acute toxicological reactions which can be safely applied in clinic.
