摘要
目的探讨CGS21680对脂多糖(LPS)诱导的小鼠大脑皮质小胶质细胞炎症细胞模型白细胞介素-1β(IL-1β)表达的影响。方法采用新生小鼠大脑皮质小胶质细胞原代培养技术,将培养的小胶质细胞随机分组,设为对照组(加入单纯的DMEM/F12完全培养基)、LPS组(制作小胶质细胞炎症细胞模型)、CGS21680组(小胶质细胞炎症细胞模型基础上叠加CGS21680处理),CGS21680组再按CGS21680不同浓度分为0.1、1和10μmol·L^(-1)CGS21680亚组。ELISA检测各组24 h后IL-1β含量。结果 CGS21680组较LPS组小胶质细胞IL-1β表达量显著下降。结论 CGS21680可明显抑制LPS诱导的小鼠大脑皮质小胶质细胞的炎症反应。
Aim To investigate the effect of CGS21680, an adenosine A_(2A) receptor agonist, on the expression of IL-1β in lipopolysaccharides-induced(LPS-induced) inflammation in microglia cells. Methods Primary murine microglia cells were cultured and randomly divided into three groups: a control group, a LPS group(established inflammation model in vitro), a LPS+CGS21680 group(inflammation model in combination with CGS21680 treatment). Twenty-four hours after the treatment, the expression of IL-1β by microglia was detected by ELISA. Results The expression of IL-1β was reduced in CGS21680 treatment group compared with LPS group. Conclusion Adenosine A_(2A) receptor agonist CGS21680 inhibits the expression of IL-1β in LPS-induced inflammation in microglia cells.
