摘要
目的:探讨热休克蛋白90α(HSP90α)在非小细胞肺癌(NSCLC)患者血清中的表达水平及其临床意义。方法:选取2015年10月至2016年10月期间,在第四军医大学唐都医院胸外科确诊为NSCLC的患者100例为肺癌组。选取同期在我院进行体检的健康成年人40例为对照组。检测并比较两组血清HSP90α表达水平差异;比较不同肿瘤分期(TNM分期)患者的血清HSP90α水平差异;探讨不同TNM分期与血清HSP90α表达水平之间的关系。培养NSCLC A549细胞,检测其分泌至胞外的HSP90α蛋白;使用HSP90α抑制剂处理A549细胞,MTT法检测细胞增殖能力的变化。结果:肺癌组与对照组患者血清HSP90α表达水平比较,差异有统计学意义(P<0.001);不同TNM分期患者血清HSP90α表达水平比较,差异有统计学意义(P<0.001);Spearman秩相关分析显示,TNM分期与血清HSP90α水平呈正相关(r=0.95,P<0.05)。体外细胞实验证实A549细胞能分泌HSP90α至细胞外,使用HSP90α抑制剂处理后细胞增殖能力明显下降。结论:NSCLC患者血清中HSP90α表达水平较正常人升高,并且与TNM分期呈正相关。血清HSP90α由肺癌细胞分泌入血,检测其水平对肺癌的诊断与治疗有一定的临床参考价值。
Objective:To measure serum levels of heat shock protein 90α(HSP90α) in patients with NSCLC and to evaluate their clinical significance.Methods:A total of 100 patients with NSCLC and 40 controls were enrolled in this study.Serum levels of HSP90α were measured by ELISA.Serum HSP90α was compared between the two groups and among patients in diferent TNM stages at the mean time.The correlativity between TNM staging and serum HSP90α were investigated.In vitro studies,we cultured the A549 cells and detected extracellular HSP90α which the A549 cells secreted,then used HSP90α neutralizing antibody processing A549 cells,to detect the change of cell proliferation by MTT method.Results:Serum levels of HSP90α were significantly higher in patients with NSCLC than in controls(P〈0.001).Patients with different TNM stages were significantly different(P〈0.001) in serum HSP90α.The Spearman rank sum test showed that TNM staging and serum HSP90α level were positively correlated(r= 0.95,P〈0.05) with each other.In vitro studies,A549 cells can secrete HSP90α,HSP90α inhibitor can decrease cell proliferation capacity significantly.Conclusion:NSCLC patients have higher levels of serum HSP90α than healthy persons.Higher TNM staging causes higher serum HSP90α levels.Serum HSP90α is secreted by NSCLC cells and can play special biological function,it can be used as a potential marker for NSCLC.
