摘要
目的探讨右美托咪定对卵巢癌细胞相关生物学行为的影响。方法通过CCK-8检测右美托咪定对卵巢癌SKOV3细胞活性的影响;通过流式细胞术FITC-Annexin V/PI双染色验证右美托咪定能否诱导SKOV3卵巢癌细胞凋亡;划痕实验检测右美托咪定对SKOV3卵巢癌细胞迁移能力的影响;Transwell侵袭实验检测右美托咪定对SKOV3卵巢癌细胞侵袭能力的影响。结果右美托咪定能降低SKOV3卵巢癌细胞的活性,且呈剂量依赖性和时间依赖性;右美托咪定能诱导SKOV3卵巢癌细胞凋亡,当右美托咪定浓度达到10、100 nmol/L,凋亡率分别为8.21%和8.30%;右美托咪定能减弱SKOV3卵巢癌细胞的迁移能力和侵袭能力。结论右美托咪定能通过诱导细胞凋亡抑制SKOV3卵巢癌细胞的活性,减弱细胞的迁移和侵袭能力。
Objective To investigate the effect of dexmedetomidine on the biological behavior of ovarian cancer cells. Methods The effect of dexmedetomidine on the activity of ovarian cancer SKOV3 cells was detected by CCK-8 kit. Flow cytometry FITC-Annexin V/PI double staining was used to test whether dexmedetomidine could in- duce the apoptosis of SKOV3 ovarian cancer cell. The effect of dexmedetomidine on the migration ability of SKOV3 o- varian cancer cells was examined by scratch healing experiment. The effect of dexmedetomidine on the invasion ability of SKOV3 ovarian cancer cells was detected by Transwell invasion assay. Results Dexmedetomidine could decreasethe activity of SKOV3 ovarian cancer cells in a dose-dependent and time-dependent manner. Dexmedetomidine could induce the apoptosis of SKOV3 ovarian cancer cells. When the concentration of dexmedetomidine reached 10 and 100 nmol/L,the apoptotic rates were 8.21% and 8.30% respectively. Dexmedetomidine could decrease the migration and invasion ability of SKOV3 ovarian cancer cells. Conclusion Dexmedetomidine can inhibit the activity of SKOV3 o- varian cancer cells by inducing apoptosis, and also can decrease its migration and invasion ability.
