摘要
目的研究绿色闪烁光诱导豚鼠近视眼模型中乙酰胆碱M1受体的表达,探讨M1受体在近视发展过程中的作用和机制。方法选择健康20日龄豚鼠30只,随机分为3组,每组10只,绿光组用明暗交替各2 s的绿色闪烁光照射,放在完全密闭的纸箱中饲养;自然光组用正常自然光照射,放在四周密闭而上方开放的纸箱中饲养;对照组用正常自然光照射,放在视野开阔的笼中饲养。分别在实验前、实验6周时用散瞳检影验光法测量眼屈光度,用A超测量眼轴长度。用逆转录聚合酶链反应(RT-PCR)方法检测视网膜、脉络膜和巩膜组织中的M1受体m RNA的表达。结果绿光组诱导出近视,眼轴长度均较自然光组和对照组增加,差异均有统计学意义(P<0.05)。绿光组巩膜、视网膜组织M1受体m RNA的表达均低于自然光、对照组,差异均有统计学意义(均P<0.01)。结论绿色闪烁光诱导的豚鼠近视可能与眼视网膜组织、巩膜组织中M1受体m RNA的表达量减少有关。
Objective To research the expression of muscarinic receptors-1 (M1) in green flashing light-induced myopia in guinea pig,and to explore the role and mechanism of M1 receptor in development of myopia. Methods Thirty heahy guinea pigs 20-day-old were randomly divided into three groups,green light group reared in tightly closed carton illuminated by green flashing light with an interval of 2 s, natural light group reared in the equally sized carton closed around with an open top illuminated by natural light,and control group reared in a sight-widen cage illuminated by natural light.After 6-week treatment, refaction was measured respectively by using retinoscopy and the ocular axial was determined by A-scan ultransonography. The mRNA expression of M1 receptor in choroids,retina and scleral was detected by RT-PCR. Results In green light group, guinea pigs developed myopia, axial length significantly increased compared with those in natural light group and the control group(P〈 0.05). Expressions of mRNA of M1 receptor in sclera and retina were lower compared with those in natural light group and control group(P〈0.01). Conclusion After stimulation by green flashing light for eight weeks, guinea pigs developed myopia and eye axial elongated.The expressions of mRNA of M1 receptor in sclera and in retina have reduced in guinea pigs which are stimulated by green flashing light.
