人参皂苷Rg5通过下调Bcl-2蛋白促进食管癌Eca-109细胞凋亡 被引量:10

Ginsenoside Rg5 Promotes Apoptosis of Esophageal Cancer Eca-109 Cells by down-regulating Bcl-2 Protein

摘要 目的研究人参皂苷Rg5对于人食管癌细胞Eca-109凋亡的影响以及相关机制。方法传代培养人食管癌细胞Eca-109,应用CCK8法检测人参皂苷Rg5对于Eca-109细胞增殖的影响。应用流式细胞仪检测人参皂苷Rg5作用后Eca-109细胞的凋亡变化。使用Western blot法检测C-caspase3、C-caspase9、C-parp、Bcl-2、Bax相关凋亡蛋白的变化情况。结果人参皂苷Rg5明显抑制Eca-109细胞的生长,同时促进细胞凋亡,且均呈现剂量依赖性。Western blot显示,随着人参皂苷Rg5浓度的增高,细胞中的C-caspase3、C-caspase9、C-parp蛋白表达增多,而Bcl-2表达明显降低。结论人参皂苷Rg5抑制人食管癌细胞Eca-109增殖并促进其凋亡,发挥该作用与Bcl-2蛋白下调有关。 Objective To study the effect of ginsenoside Rg5 on the apoptosis of human esophageal cancer cell line Eca-109 and related mechanisms.Methods The human esophageal cancer cell line Eca-109 was subcultured and the effect of ginsenoside Rg5 on the proliferation of Eca-109 cells was detected by CCK8 assay.Flow cytometry was used to detect the apoptosis of Eca-109 cells induced by ginsenoside Rg5.Western blot was used to detect the changes of Cparp3,C-caspase9,C-parp,Bcl-2 and Bax related apoptotic proteins.Results Ginsenoside Rg5 significantly inhibited the growth of Eca-109 cells and promoted cell apoptosis in a dose-dependent manner.Western blot showed that the expression of C-caspase3,C-caspase9 and C-parp increased and the expression of Bcl-2 decreased significantly with the increase of ginsenoside Rg5 concentration.Conclusion Ginsenoside Rg5 can inhibit the proliferation and promote the apoptosis of human esophageal cancer cell line Eca-109.This effect is related to the down-regulation of Bcl-2 protein.
