摘要
目的探讨磷酸二酯酶4D交互蛋白(PDE4DIP)基因对布加综合征(BCS)隔膜形成的影响。方法通过小干扰RNA特异性沉默人血管内皮细胞(HUVECs)中的PDE4DIP基因,应用CCK8实验、划痕实验及血管形成实验分别检测转染后各组的增殖、迁移及血管形成能力,分析其改变情况。结果沉默该基因后,其mRNA及蛋白水平表达均下降,差异均有统计学意义(P〈0.05);各组细胞增殖的差异无统计学意义(P=0.51),迁移能力(P=0.003)及血管形成能力(P=0.001)明显降低。结论在HUVECs细胞中,PDFADIP基因沉默具有抑制HU—VECs细胞迁移及血管形成能力的作用,可能与BCS隔膜形成有关。
Objective To investigate the relationship between phosphodiesterase 4D interacting protein (PDE4DIP) gene and Budd- Chiari syndrome (BCS). Methods The sinai1 interference RNA (siRNA) targeting PDFADIP gene and the controls were transfected into human umbilical vein endothelia cells (HUVECs) , respectively. Then the CCK8 assay, migration scratch assay and tube formation assay were applied to detect the effects on cell proliferation, migration and angiogenesis. Results The expression of PDE4DIP mRNA and protein cells were decreased after interference (P 〈0. 05 ). After PDFADIP gene silencing, no remarkable changes were found in the proliferation of each group ( P = 0. 51 ). PDFADIP gene silencing could markedly decrease the ability of migration ( P = 0. 003 ) and angiogenesis ( P = 0. 001 ). Conclusions PDFADIP gene silencing can inhibit the ability of migration and angiogenesis in HUVECs cells, which may be associated with the membrane formation of BCS.
