猪血清和血浆外泌体miRNA的鉴定与比较 被引量:1

Identifying and comparing the exosome miRNA spectrum between porcine serum and plasma

摘要 为了解血液中外泌体miRNA的表达情况,利用高通量测序技术检测了来源于相同个体的猪血清和血浆外泌体miRNA表达谱。结果显示,血清外泌体中提取的RNA浓度显著高于血浆外泌体(P=0.02),从2个文库中共检测到187个成熟体m-RNAs,对应165个前体。鉴定出了在2个文库中丰富表达的17个miRNAs,暗示这些miR—NAs在血清和血浆中发挥着重要的作用。筛选出99个差异表达miRNAs,其中54个在血清外泌体中高表达,45个在血浆外泌体中高表达。通过靶基因预测和功能注解分析,丰富表达的miRNAs广泛地参与到机体的生理学过程中。本研究对猪血清和血浆外泌体miRNA进行了鉴定及比较,为深入研究外泌体miRNA在生物体内的功能提供理论依据。 Detection of er,diabetes and other b d ood biomarkers is an effective method seases. At present, exosome miRNAs attention by researchers. In order to identify the expression for diagnosis and treatment in canc- as biomarkers have been paid more of miRNAs in blood exosomes, this study explored porcine serum and plasma exosomes miRNA expression profiling by high-through- put sequencing technology. Totally 187 mature miRNAs corresponding to 165 precursors were i- dentified in both libraries,and the concentration of serum exosomes was significantly higher than that of plasma exosomes(P=0.02). 17 abundant miRNAs in the two libraries were found,sugges- ting that these miRNAs play an important role in both serum and plasma. Of the 99 differentially expressed miRNAs,54 were highly expressed in the serum exosomes, and the rest 45 were highly expressed in plasma exosomes. According to target gene prediction and functional annotation anal- ysis,miRNAs with abundant expression levels were involved in a wide range of physiological processes in the whole body. Our study identifying and comparing the exosome miRNA between porcine serum and plasma provides a preliminary basis for further study on the function of exo- some miRNA in vivo.
