摘要
目的优化酶法提取土茯苓多糖的工艺。方法通过单因素试验法和响应面分析法,考察最佳酶的酶解时间、酶解p H、酶用量对提取土茯苓多糖的影响。结果木瓜蛋白酶提取多糖效率最高,其最佳工艺为酶用量0.6%,酶解p H为8,酶解时间150 min,在此条件下得到的实际多糖提取率为26.31%,与预测值25.71%的相对误差为2.33%。说明采用响应面分析法优化木瓜蛋白酶提取土茯苓多糖的方法可靠。结论此设计法用于酶法提取土茯苓多糖可行。
Objective To optimize the extraction technique of polysaccharide from Rhizoma Smilacis Glabrae by enzymatic method.Methods On the basis of single factor experimental and response surface methodology to investigate the influence of enzymatic time p H and dosage on extraction yield of polysaccharide.Results The optimum enzymatic was papain and its optimum conditions were as follows:enzyme concentration of 0.6%,enzymatic hydrolysis PH of 8,enzymatic hydrolysis time of 150 min,the enzymatic hydrolysis temperature of 50℃.Under these conditions above,the actual extraction ratio of polysaccharides from Rhizoma Smilacis Glabrae was26.31%,which was well matched with the predicted value(25.71%).Conclusion This design method to extract polysaccharide from Rhizoma Smilacis Glabrae by enzymatic is feasible.
