制备氧化石墨烯及其与鱼精DNA的作用 被引量:1

摘要 本文以氧化还原法合成制备氧化石墨烯,用原子力显微镜对其尺寸及厚度进行表征。其作为基底材料,通过酰胺反应偶联核酸分子探针,从而建立高效、高灵敏度的纳米生物传感器。文中探究了鱼精DNA种类对探针工作的影响,发现50bp的鱼精DNA可实现较好的氧化石墨钝化作用,提升检测信号。该生物传感器可对pH具有良好的响应,且可在复杂环境下稳定存在。这些优点使得基于氧化石墨烯构造的纳米生物传感器能够实现对pH的高灵敏检测,且具有良好的稳定性,为氧化石墨烯在生物医学上的应用提供了重要依据。 The graphene oxide was prepared by traditional redox method and characterized by AFM. Through the amidation reaction between graphene oxide and amino-DNA probe, we synthesized a pH sensitive nanoprobe with higher efficiency and sensitivity. The report studied the influence of herring sperm DNA(HSD) incubation during the preparation process, and found that 50bp HSD was the best for passivating the surface of GO to enhance probe signal. Simultaneously, pH probe can be protected by graphene oxide with enhanced stability which allows the pH detection under complex environment. The mentioned merits of nano-biosensor constructed from graphene oxide conjugated DNA probe provide a wide application potential in biomedicine for the graphene oxide.
