摘要
目的:测定不同产地雾水葛中槲皮素的含量。方法:本实验采用HPLC法测定药材雾水葛中槲皮素的含量。流动相甲醇-0.1%磷酸溶液(53︰47),检测波长254 nm,流速1.0 m L/min,柱温25℃。结果:在此方法下,雾水葛中槲皮素得到较好分离,其线性范围为0.016~0.080mg/m L(r=0.99905),平均加样回收率为100.07%(n=9)。药材雾水葛中槲皮素含量差异较大,槲皮素的含量分别为:广州:0.1101 mg/g;中山:0.2585 mg/g;河源:0.2972 mg/g;肇庆:0.2640 mg/g;韶关:0.3230 mg/g;汕头:0.0846 mg/g。结论:该方法操作简便,结果准确,重复性好,可为该药材合理开发利用提供科学依据。
Objective To establish a method for the determination of Quercetin in Pouzolzia zeylanica (L.) Benn from different areas. Methods The contents of Quercetin were determined by HPLC. The mobile phase consisted of methanol-0.1% phosphoric acid (53 : 47), the detection wavelength was 254 nm, with the flow rate of 1.0 mL/min and the temperature at 25℃. Results Under the chromatography conditions, quecertin was separated effectively from other compounds existing in the extract of Pouzolzia zeylanica(L.)Benn. The results showed that the method was to proved to be good linear relationship in the range of 0.016 mg/mL -0.080 mg/mL(r =0.99905)for quercetin. The average recoveries of quecertin were 100.07 %.The contents of Quercetin in Pouzolzia zeylanica (L.) Benn. from different areas varied greatly. The contents of quercetin in Pouzolzia zeylanica(L.)Benn were as follow: Guangzhou: 0.1101 mg/g Zhongshan: 0.2585 mg/g; Heyuan: 0.2972 mg/g;Zhaoqing: 0.2640 mg/g: Shaoguan: 0.3230 mg/g;Shantou: 0.0846 mg/g. Conclusion: The method for determination of Quercetin is simple, accurate and with good repeatability, which could provide a scientific basis for rational development and utilization.
