IL4和IL6基因多态性与胃癌风险的相关性研究

Associations of Genetic Polymorphisms in IL4 and IL6 with Risk of Gastric Cancer

目的研究IL4和IL6基因多态性与胃癌风险的相关性。方法选择2016年6月至2017年6月于陕西中医药大学第二附属医院就诊的符合入选条件的胃癌患者共300例(病例组),以及健康对照人群300例(对照组),分别采集血样。采用Mass ARRAY方法对rs2243250、rs2227284、rs2243267、rs1800796、rs2069837和rs2069840六个位点进行基因分型,用SPSS 19.0在线软件计算基因多态性与胃癌患病风险的相关性。结果 6个候选SNP位点全都符合哈德-温伯格平衡(P-HWE>0.05),通过比较病例组和对照组的最小等位基因频率发现,rs1800796和rs2069837的最小等位基因G与胃癌风险增加具有相关性(P<0.05)。通过比较两组间的基因型频率差异发现,rs2243267和rs1800796的GC和GG基因型,rs2069837的AG和GG基因型,均与胃癌患病风险增加相关(P<0.05)。引入遗传模型分析后发现,rs2243267、rs1800796和rs2069837在三个遗传模型下都与胃癌风险增加具有相关性(P<0.05);而rs2069840仅在显性模型下与胃癌风险增加具有相关性(P<0.05)。此外,单体型分析显示由rs1800796、rs2069837和rs2069840三个位点组成的GAC单体型与胃癌患病风险增加具有相关性(P<0.05)。结论 IL4基因rs2243267位点和IL6基因的rs1800796、rs2069837、rs2069840位点以及其组成的单体型GAC可能与胃癌风险增加具有相关性。

Objective To investigate the association between single nucleotide polymorphisms（ SNPs） in IL4 and IL6 and the risk of gastric cancer（ GC）.Methods A total of 300 GC patients and another 300 healthy controls were enrolled in the Second Affiliated Hospital of Shaanxi University of Chinese Medicine from June 2016 to June 2017. Blood samples were collected. The genotypes of rs2243250,rs2227284,rs2243267,rs1800796,rs2069837 and rs2069840 were determined with Mass ARRAY,and the association of SNPs with the risk of GC was evaluated with SPSS 21.0. Results All the six candidate SNPs conformed to the Had-Weinberg equilibrium（ P-HWE〉0.05）. By comparing the allele frequency of the patients and controls,we found that the allele G of rs1800796 and rs2069837 was associated with an increased risk of GC（ P〈0.05）. By comparing the difference in genotype frequency between the two groups,it was found that the GC and GG genotypes of rs2243267 and rs1800796,the AG and GG genotypes of rs2069837 were all associated with an increased risk of GC（ P 〈0. 05）. Genetic model analysis suggested that rs2243267,rs1800796 and rs2069837 were associated with an increased risk of GC in all the three genetic models（ P〈0.05）,while rs2069840 was only associated with an increased risk of GC in the dominant model（ P〈 0.05）. In addition,haplotypes analysis showed that GAC haplotype composed of rs1800796,rs2069837 and rs2069840 was associated with an increased risk of GC（ P〈0.05）. Conclusion Rs2243267 of IL4,rs1800796,rs2069837 and rs2069840 of IL6,and its haplotype GAC may be correlated with the increased risk of GC.
