摘要
目的制备多羧基笼型倍半硅氧烷(POSS-COOHx)杂化硅橡胶(SR),并研究其生物相容性。方法 (1)八苯基倍半硅氧烷经过卤代反应和氧化还原反应得到多羧基八苯基倍半硅氧烷,而后与液态双组分SR混合均匀硫化得POSS-COOHx杂化SR。(2)采用CCK-8和细胞骨架染色检测细胞相容性;通过体内置入实验和HE染色,检测POSS-COOHx杂化SR的生物相容性。结果 (1)成功制备POSS-COOHx杂化SR。(2)POSS-COOHx杂化SR的生物相容性较普通SR显著提高。结论笼型倍半硅氧烷可以作为很好的分子平台引入亲水性基团改变SR的表面性质,提高SR的生物相容性。
Objective To prepare multicarboxylsilsesquioxane (POSS-COOHx) hybrid silicone rubber and study its biocompatibility. Methods (1)Through halogenation and oxidation reduction reaction, eight phenyl silsesquioxane was converted to muhicarboxyl eight phenyl silsesquioxane, and was then mixed with liquid two-component silicone rubber, the mixture was cured at room temperature and muhicarboxylsilsesquioxane (POSS-COOHx) hybrid silicone rubber was acquired. (2)Cell compatibility was examined by CCK-8 and cell skeleton staining. In vivo implantation test and HE staining were used to detect the biocompatibility of muhicarboxylsilsesquioxane (POSS-COOHx) hybrid silicone rubber. Results (1)Muhicarboxylsilsesquioxane (POSS-COOHx) hybrid silicone rubber was successfully prepared; (2)The biocompatibility of mnhicarboxylsilsesquioxane (POSS-COOHx) hybrid silicone rubber improved more significantly than ordinary silicone rubber. Conclusion Cage-type silsesquioxane can be used as a good molecular platform to introduce hydrophilic groups to change the surface properties of biomaterials and to improve the biocompatibility of biomaterials.
