摘要
用成年新西兰大白兔构建A,B两类关节软骨缺损动物模型。A模型为未钻通软骨下骨模型,采用同体对照,右侧为(种子细胞+白及胶+PluronicF-127)复合支架材料修复;左侧为空白对照。B模型为钻通软骨下骨模型,同体对照,右侧为(元细胞白及胶+PluronicF-127)复合支架材料修复;左侧为空白对照。另设一组采用B模型动物,用PluronicF-127人工合成材料修复,双侧相同。在术后4,12,24周处死动物,取修复区组织作HE染色、Ⅱ型胶原免疫组化染色、阿新蓝染色和甲苯胺蓝染色,光镜下观察,参照Wakitanis组织学评分标准,对各组动物修复组织形态学进行半定量评分。A,B2组动物软骨缺损模型,均为透明软骨修复,结果满意。无细胞白及胶与PluronicF.127可注射型复合支架材料,与有种子细胞+自及胶+Plu-tonicF-127复合物支架材料,二者对关节软骨缺的损修复效果无明显差异,有白及胶的人工合成材料优于单纯人工合成材料的修复,提示白及胶在复合支架材料修复关节软骨缺损中起了莺要作用。
Two types (A model and B model) of articular cartilage defect models were prepared by using adult New Zealand white rabbits. A model group was applied by drilling without through subchondral bone, whose right joint was repaired by composite scaffolds made by seed cell, gum-bletilla as well as Pluronic F-127, and left side was blank control: B model group was applied by subchondral drilling method, whose right joint was repaired by using composite scaffolds made by gum-bletilla and Pluronic F-127 without seed cells, and left side was blank control. Autogen0us contrast was used in both model types. In addition, another group was applied with B model type rabbits, which was repaired with artificial complex material of Pluronic F-127 in both joint sides. 4, 12 and 24 weeks af- ter operation, the animals were sacrificed and the samples were collected from repaired area for staining with HE, type Ⅱ collagen im- munohistochemical method, AIcian blue, and toluidine blue, and then were observed with optical microscope. Semi-quantitative scores were graded by referring to Wakitanis histological scoring standard to investigate the histomorphoiogy of repaired tissue. Hyaline carti- lage repairing was achieved in both Group A and Group B, with satisfactory results. There were no significant differences on repairing effects for articular cartilage defects between composite scaffolds made by seed cell, gum-bletilla and Pluronic F-127, and the compos- ite scaffolds made by gum-bletilla and Pluronic F-127 without seed cell. Better repairing effects for articular cartilage defects were ob- served in groups with use of gum-bletilla, indicating that gum-bletilla is a vital part in composite scaffolds material.
