跑台运动对衰老大鼠免疫功能的实验研究 被引量:3

摘要 目的:探讨6周跑台运动对D-半乳糖致衰老模型大鼠免疫功能的影响,以此阐明运动延缓衰老的部分机制。方法:将雄性SD大鼠随机分为正常对照组(NC)组,衰老对照(AC)组,衰老训练(AE)组。AC和AE组大鼠连续腹腔注射125mg/kg D-半乳糖,1次/d,NC组注射等量的生理盐水,持续6周。同时,AE组进行6周中小强度跑台训练。观察各组大鼠的生存状态及生物学行为,并定期监测其体重变化。测定衰老大鼠脾脏指数及观察其脾脏结构显微形态学改变;大鼠衰老模型的鉴定采用免疫组织化学法检测衰老相关蛋白P16蛋白、β-半乳糖苷酶表达;ELISA检测大鼠血清免疫球蛋白IgG、血清IL-2含量变化;免疫组织化学法检测脾脏组织IL-2的表达情况。结果 :与AC组比,AE组大鼠脾脏指数,脾脏形态结构,衰老细胞SA-β-Gal阳性率、P16蛋白表达情况,大鼠血清IgG、血清IL-2含量以及脾脏组织IL-2阳性细胞表达均得到明显改善,差异具有统计学意义(P<0.05或P<0.01)。结论 :6周中小强度跑台训练在一定程度上可以延缓机体免疫功能的增龄性衰退,其机制与运动显著提高机体IL-2的表达有关。 Objective:To explore the effect of 6-week treadmill exercise on immunlogic function of aging rats which influenced byD-galactose.Method:Twenty-four male SD rats were randomly divided into normal control group(NC),aging control group(AC)and aging exercise group(AE).The AC and AE group were continually injected with D-galactose 125 mg/kg,qd x 42 days.The NC group were treated with equal amout of saline at the same time.The AE group participated in 6-week treadmill exercise at moderate intensity.To observe aging rats' living state and biological behavior,recording their body weight changes.To detect spleen index and observe spleen microscopic structures.Aging rats model was identified by protein P1 6 andβ-galactosidase which detected by immune histochemical methodon.Using ELISA method to detected the level of IgG and IL-2 in serum.IL-2 protein expression of splenocytes were detected by immune histochemistry.Results:Compared with AC group,the spleen index,changes in spleen microscopic structures,the expression of P16 protein and the rate of SA-β-Gal staining positive cells of AE group were improved obviously.The level of IgG and IL-2 in serum of AE group were increased obviously compared with AC group(P0.05 or P0.01).Conclusion:6-week treadmill exercise can delay the age related decline of immune function.Its mechanism is related to the improvement of IL-2.
