响应面法优化杜仲叶中绿原酸提取工艺 被引量:24

Optimization of extraction process of chlorogenic acid in Eucommia ulmoides leaves by response surface methodology

摘要 为了优化杜仲叶中绿原酸的提取工艺,采用有机溶剂萃取杜仲叶中的绿原酸,研究乙醇质量分数、固液比、提取温度、提取时间、提取次数等因素对绿原酸提取率的影响。在单因素试验的基础上优选试验因素与水平,根据中心组合试验设计原理,采用3因素3水平响应面分析法,拟合实验因素与响应值的多元二次方程,并对各因素的显著性及交互作用进行分析,运用Design-Expert 8.0 Trial软件优化得到提取杜仲叶中绿原酸的工艺最佳参数。结果表明:在料液比1∶11、乙醇质量分数40%、提取温度61.4℃的条件下,绿原酸提取率为0.872 8%。 In order to optimize extraction process of chlorogenic acid from Eucommia ulmoides leaves,chlorogenic acid in E.ulmoides leaves was extracted in organic solvent,and effects of ethanol mass fraction,solid-liquid ratio,extraction temperature,extraction duration,extraction times and other factors on extraction rate of chlorogenic acid were researched.On the basis of single factor experiment,experiment factors and levels were selected,according to the central composite experimental design principle,using 3 factors and 3 levels of response surface analysis method,a multivariate quadratic equation of the experimental factors and response values was fitted,significances of different factors and their interactions were analyzed,and the optimal parameters of the extraction process of chlorogenic acid in E.ulmoides leaves were optimized by using Design-Expert 8.0 Trial software.The results show that:under the conditions of solid-liquid ratio 1 ∶ 11,ethanol mass fraction 40% and extraction temperature 61.4 ℃,the extraction rate of chlorogenic acid was 0.872 8%.
