星点设计-效应面法优化过墙风总黄酮的提取工艺

摘要 目的:优化瑶药过墙风总黄酮提取工艺,为其开发和利用提供参考。方法:在单因素试验筛选基础上,以乙醇浓度、提取温度和提取时间为自变量,总黄酮得率为因变量,然后根据星点设计原理进行试验,对结果进行多元线性回归和二项式方程拟合,采用效应面法优化过墙风总黄酮的提取工艺。结果:过墙风总黄酮最优提取工艺为乙醇浓度54.76%,提取温度83.92℃,提取时间102.64 min,固液比为1∶20,提取2次。结论:本实验星点设计-效应面法优化过墙风总黄酮提取工艺简便可行,预测结果可靠,适于工业化生产。 Objective:To optimize the extraction process of total flavonoids from Clerodendrum philippinum Schauer var.simplex Mlodenke roots.Methods:On the basis of single-factor tests,experiments were performed according to central composite design with flavonoids extraction percentage as dependent variable,and the extracting temperature,extracting time and ethanol concentration as independent variables.The results were investigated by multiple linear regression and binomial fitting,and the response surface method was employed to predict the optimal formulation of extraction process.Results:The optimization of extraction process were ethanol concentration 54.76%,extraction temperature 83.92 ℃,extracting for 102.64 min,solid-liquid tatio of 1∶20 and extracting for twice.The extraction yield of total flavonoids was 5.021%.Conclusion:The extraction process of total flavonoids from Clerodendrum philippinum Schauer var.simplex Mlodenke roots optimized by central composite design-response surface method was simple,highly predictive,and suitable for industrialized production.
