TGF-β/SMAD信号通路与EMT及肺纤维化关系的研究进展 被引量:13

摘要 在肺纤维化发病过程中,多条SMAD依赖及非SMAD依赖信号通路参与上皮-间充质细胞转分化(EMT)及纤维化的形成和发展,其中转化生长因子-β(TGF-β)/SMAD信号通路在EMT的形成及信号通路间的联系中发挥着关键性的作用。在该通路中,TGF-β与受体结合后,激活下游的SMAD蛋白,活化的SMAD2与SMAD3形成同源或异源二聚体后与SMAD4共同进入细胞核,与纤维化相关转录因子相互作用,共同调节靶基因的沉默或激活,从而促进EMT及肺纤维化的形成。本文对该通道与EMT及肺纤维化的关系作一综述。
