基于Miseq测序技术分析不同元胡产区土壤细菌功能基因组成与差异 被引量:8

Analysis of Composition and Difference of Soil Bacteria Functional Genes of in Different Corydalis Yanhusuo Producing Areas Based on Miseq Sequencing Technology

摘要 [目的]分析元胡道地产区和非道地产区根际土壤细菌功能基因的组成和差异,在基因层面上探究根际土壤细菌对元胡生长的影响。[方法]将同一种源的元胡分种在不同产区,在收获期采集其根际土,Miseq测序检测元胡根际微生物及功能基因组成,与COG以及KEGG数据库进行比对。[结果]陕西非道地产区和浙江道地产区根际细菌组成种类相似,组成丰度在优势菌中差异明显:门水平上酸杆菌门、厚壁菌门浙江组较高,变形菌门、拟杆菌门浙江组较低;属水平上酸杆菌属Gp1浙江组较高,鞘氨醇单胞菌属、疣微菌某属浙江组较低。根际细菌功能基因经COG及KEGG数据库分析,分别被注释到24个功能分类和41个信号通路中,基于COG数据库的24个功能分类差异分析发现,陕西非道地产区和浙江道地产区的元胡根际细菌功能基因在RNA加工和修饰、染色质结构和动力学、糖类运输和代谢等15个分类中均存在差异;基于KEGG数据库的41个信号通路差异分析发现,陕西非道地产区和浙江道地产区的元胡根际细菌功能基因在糖代谢、氨基酸代谢、细胞生长等21个通路中存在差异。基于功能的聚类分析、主成分分析及NMDS分析显示道地和非道地产区元胡根际细菌土壤功能基因组成呈现明显地域差异,陕西三组聚为一类,浙江三组聚为一类。[结论]元胡土壤细菌功能基因组成在道地和非道地产区间差异较大,功能基因组成与元胡生长密切相关,与药材道地性的产生也可能具有相关性。 [Objective]To analyze the composition and differences of rhizosphere soil bacterial functional genes of Corydalis yanhusuo in the new and genuine producing areas,and to explore the effects of rhizosphere soil bacteria on the growth of C.yanhusuo at the genetic level.[Methods]C.yanhusuo was planted in different producing areas from the same provenances,rhizosphere soil was harvested at the harvesting stage.The rhizosphere microbes and functional gene compositions were detected by Miseq sequencing and compared with COG and KEGG databases.[Results]The composition of rhizosphere bacteria was similar in the new producing areas of Shaanxi Province and genuine producing areas of Zhejiang Province and the abundance of the dominant species had significant differences.On the level of Phylum,the content of Acidobacteria,Firmicutes in Zhejiang was higher than in Shaanxi.In contrast,the content of Proteobacteria,Bacteroidetes in Zhejiang was lower.On the level of Genus,the content of Gp1 in Zhejiang was higher than them in Shaanxi.In contrast,the content of Sphingomonas,Subdivision 3 genera in Zhejiang was lower.The functional genes were annotated to 24 functional categories and 41 signaling pathways respectively by COG and KEGG databases.There were 15 functional categories including RNA processing and modification,chromatin structure and dynamics and carbohydrate transport and metabolism having differences in the fresh and original producing areas based by COG databases.There were21 signaling pathways including sugar metabolism,amino acid metabolism and cell growth having differences by KEGG databases.The analysis based on the functional principal components and NMDS also showed regional difference of rhizosphere soil bacterial functional genes 'composition of C.yanhusuo in the fresh and original producing areas.[Conclusion]The bacterial functional genes' composition of C.yanhusuo soil are quite different in new and original producing areas.The composition of functional genes is closely related to the growth of C.yanhusuo,and may also be related to the originality of herbs.
