摘要
目的为了解吉林省2009年-2016年肠道病毒71型(EV71)流行株的基因特征。方法对吉林省2009年-2016年手足口病来源的EV71流行株进行VP1编码区逆转录PCR反应(RT-PCR)扩增,并对扩增产物进行核苷酸序列测定,使用Mega 6.0和Bioedit 7.01软件对扩增序列进行基因亲缘关系和氨基酸位点变异分析。结果吉林省2009年-2016年的97株EV71流行株与C4a基因亚型参考株位于同一簇支上,均为C4a基因亚型;97株EV71流行株在VP1编码区共有25处氨基酸位点变异,其中流行株突变数量较多的位点为22氨基酸位点和289氨基酸位点。结论吉林省2009年-2016年的97株EV71流行株均为C4a基因亚型,而且发生了多位点的氨基酸变异。
Objective To understand the gene characteristics of enterovirus 71 (EV71) in Jilin Province during 2009 -2016. Methods VPI coding region reverse transcription PCR(RT PCR) amplification was performed on EV71 prevalent strains of hand foot mouth disease from 2009 to 2016 in Jilin Province. The nucleotide sequence of the amplified products was determined using Mega 6.0 and Bioedit 7.01. The software performs genetic analysis and amino acid locus variation analysis on the amplified sequences. Results All of the 97samples belonged to C4a subtype, and 25 amino acid sites mutated in the VP1 coding region of 97 EFT1 strains. Among them, 22 amino acid site and 289 amino acid site were with the most mutant number in epidemic pathogens . Conclusion 97 strains of EFT1 epidemic in Jilin during 2009 -2016 were all C4a subtype, and amino acid site have mutated.
