摘要
目的探讨脊髓小脑共济失调症相关蛋白3(Ataxin-3)与基质金属蛋白酶-11(MMP-11)在结直肠癌中的表达及与临床病理特征的关系。方法选取2015年1月至2017年1月间粤北人民医院收治的80例结直肠癌患者的肿瘤组织标本为观察组,距癌组织>5cm的癌旁正常组织标本为对照组。采用免疫组化法检测两组组织中Ataxin-3和MMP-11的表达,分析Ataxin-3和MMP-11的表达与临床病理特征之间的关系及两者表达的相关性,采用ROC曲线分析Ataxin-3和MMP-11表达水平对结直肠癌的诊断价值。结果观察组标本Ataxin-3阳性率显著低于对照组,MMP-11阳性率显著高于对照组,差异均有统计学意义(均P<0.05)。观察组标本Ataxin-3和MMP-11表达与年龄、性别和肿瘤直径无关(P>0.05),与淋巴结转移、Dukes分期和脉管浸润有关(P<0.05)。Pearson相关性分析显示,Ataxin-3和MMP-11在结直肠癌组织中的表达呈负相关(P<0.05)。ROC分析表明,Ataxin-3和MMP-11的敏感度(Se)分别为0.83和0.90,特异度(Sp)分别为0.83和0.69,AUC值分别为0.89和0.85。结论在直肠癌组织中,MMP-11呈现高表达,Ataxin-3呈现低表达,两者呈负相关关系。Ataxin-3和MMP-11的表达与淋巴结转移、Dukes分期及脉管浸润相关,两者具有较高的诊断价值。
Objective To investigate the expression of spinocerebellar ataxia associated protein 3( Ataxin-3) and matrix metalloproteinase-11( MMP-11) in colorectal cancer and its relationship with clinicopathological features. Methods Tissue specimens were collected from 80 patients with colorectal cancer who were treated at Yuebei People's Hospital from January 2015 to January 2017 were selected as the observation group and the normal paracancerous tissues over 5 cm away from the cancerous tissues were collected as the control group. The expressions of Ataxin-3 and MMP-11 in colorectal cancer tissues and normal paracancerous tissues were detected by immunohistochemistry. The relationship between the expression of Ataxin-3 and MMP-11 and clinicopathological features were analyzed and the relationship between the expressions of Ataxin-3 and MMP-11 in colorectal cancer tissues was analyzed by Pearson correlation analysis.ROC curve was used to analyze the value of detection of the expressions of Ataxin-3 and MMP-11 in the diagnosis of colorectal cancer. Results The positive rate of Ataxin-3 was significantly lower in the observation group than that in the control group,but the positive rate of MMP-11 was significantly higher than that in the control group( P〈0. 05). The expression of Ataxin-3 and MMP-11 in the observation group was not related to age,gender and tumor size( P〉0. 05) but related to lymph node metastases,Dukes stages and vascular invasion( P〈0. 05). Pearson correlation analysis showed that there was a negative correlation between the expressions of Ataxin-3 and MMP-11 in colorectal cancer( P〈0. 05). ROC analysis showed that the sensitivity( Se) of Ataxin-3 and MMP-11 was 0. 83 and 0. 90,respectively,the specificity( Sp) was0. 83 and 0. 69,and the AUC value was 0. 89 and 0. 85,respectively. Conclusion The expression of MMP-11 is highly expressed and Ataxin-3 is lowly expressed in colorectal cancer. There is a negative correlation between the two of them. The expression of Ataxin-3 and MMP-11 is significantly correlated with lymph node metastasis,Dukes staging and vascular invasion,and both of them have high diagnostic value.
