摘要
目的观察膝痹膏摩法对膝骨性关节炎兔血清白细胞介素6(IL-6)、白细胞介素1β(IL-1β)、肿瘤坏死因子-α(TNF-α)水平的影响。方法选取8月龄左右健康的新西兰大白兔60只,随机分为正常组,模型组,手法组,膝痹膏高浓度、中浓度、低浓度组,每组10只,除正常组外均采用石膏铁丝固定法制备膝骨性关节炎模型,造模8周后膝痹膏3个浓度组给予膝痹膏摩法干预,手法组给予单纯推拿手法,空白组和模型组不干预;干预8周处死动物,取血清利用ELISA试剂盒检测IL-6、IL-1β、TNF-α。结果膝痹膏高浓度、中浓度、低浓度和手法组都能降低膝痹兔血清中的IL-6、IL-1β和TNF-α浓度,且膝痹膏中浓度组IL-6、IL-1β和TNF-α浓度含量明显低于模型组(P=0.000)。结论膝痹膏中浓度配合推拿手法即"膝痹膏摩法"可以有效地降低IL-6、IL-1β和TNF-α浓度,抑制膝骨性关节炎软骨基质降解和关节软骨的破坏,从而延缓关节软骨的退变。
Objective To observe the Xi Bi Ointment has the effect on serum levels of interleukin 6(IL-6),interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)in knee osteoarthritis rabbits.Methods A total of 60 New Zealand white rabbits,aged about 8 months,were randomly divided into normal group,model group,manipulation group and Xi Bi cream group in high,medium and low concentration groups,with 10 in each group.The model of knee osteoarthritis was established by fixed method.After 8 weeks of modeling,the Xi Bi Ointment 3 groups were given Xi Bi Ointment intervention,the manipulation group was treated by the simple massage method,the blank group and the model group were not intervened;Serum was collected to detect IL-6,IL-1β and TNF-α by ELISA kit.Results The concentration of IL-6,IL-1β and TNF-α in Serum were significantly decreased in Xi Bi Ointment group at high,medium,low.The medium group was significantly lower than the model group on IL-6,IL-1β and TNF-α(P = 0.000).Conclusion The medium ointment concentration in conjunction with the massage method that "Xi BiOintment and massage" can effectively reduce IL-6,IL-1β and TNF-α concentration,inhibition of knee osteoarthritis cartilage matrix degradation and destruction of articular cartilage,thus delaying the degeneration of articular cartilage.
