摘要
目的异常黑胆质成熟剂总黄酮ASMq对白血病K562/ADR多药耐药逆转作用机制研究及其对耐药株m RNA表达的影响。方法不同浓度的ASMq联合不同浓度的ADR作用于K562/ADR细胞株,用MTT法检测ASMq对K562/ADR的细胞毒活性及对耐药细胞株的抑制率,用RT-PCR方法分析对K562/ADR细胞ABCB-1及ABCC-1和BCL-2表达的影响。结果异常黑胆质成熟剂总黄酮ASMq对K562/ADR细胞株增值表现为抑制作用,对K562/ADR细胞耐药基因ABCC-1、ABCB-1及BCL-2同样表达抑制作用。结论异常黑胆质成熟剂总黄酮通过抑制ABCB-1、ABCC-1、BC-L2基因表达,发挥对K562/ADR细胞株耐药逆转的作用。
Objective The study on the mechanism of Abnormal Savda Munzip(ASMq)of drug resistance reversal of K562/ADR and its effect on the mRNA expression of drug-resistant strains.Methods Different concentrations of ASMq and different concentrations of ADR were given cutured K562/ADR cell lines,the cell toxicity of K562/ADR and inhibition rate of resistant cells were detemined by MTT method.The effects of the expression of ABCC-1,ABCB-1,BCL-2 in K562/ADR cells were analyzed by RT-PCR.Results ASMq has been found to down-regulater K562/ADR cell apoptosis related proteins expression:ABCC-1,ABCB-1.BCL-2 and restrain cell proliferation with dose-dependent Conclusion ASMq can reverse the drug resistance against K562/ADR cells,down-regulate the apoptosis related mRNA.
