香茶藨子茎尖愈伤组织诱导及植株再生 被引量:2

摘要 试验以香茶藨子茎尖为外植体,建立了愈伤组织诱导及植株再生体系。结果表明,诱导愈伤组织的最佳培养基为MS+NAA 0.5 mg/L+2,4-D 0.8 mg/L+6-BA 1.0 mg/L,诱导率为87.0%,愈伤呈紧密块状,生长良好;愈伤组织分化最佳培养基为MS+KT 1.5 mg/L+IBA 0.3 mg/L,愈伤分化率为55.53%;以1/2MS基本培养基添加IBA 0.5 mg/L,最利于生根培养。该体系的建立为香茶藨子规模化生产及遗传转化提供了技术平台。 Plantlet shoot tip in vitro ofRibes odoratum Wendl. were used as the explant. Established a stable and high efficient callus induction and plants regeneration system. The results showed that the best medium for callus induction was MS medium supplementing with NAA 0.5 mg/L, 2,4-D 0.8 mg/L and 6-BA 1.0 mg/L, callus induction rate was 87.0%, callus compact and well developed; the optimal callus differentiation medium was MS upplemented with KT 1.5 mg/L and IBA 0.3 mg/L, with callus differentiation rate up to 55.53%; the proper rooting culture was 1/2MS upplemented with IBA 0.5 mg/L. These results can provide the technical basis for scale production and genetic transformation of plantlets of Ribes odoratum Wendl..
