摘要 目的:探讨新型矿化胶原膜(the mineralized collagen membrane)对人成骨肉瘤细胞株Saos-2生物学行为的影响。方法:将人成骨肉瘤细胞Saos-2接种于矿化胶原膜表面,光镜观察细胞形态;制备胶原膜的浸提液,以正常培养基为阴性对照,Bio-gide的浸提液为阳性对照,检测三组细胞的增殖和碱性磷酸酶(ALP)的活性。采用SPSS17.0软件对数据进行统计学分析。结果:矿化胶原膜和Saos-2细胞共培养时,细胞伸展良好;随着培养时间的延长,对照组和浸提液组细胞数量不断增加,1、4、7d时三组OD值之间无显著性差异,10d时阴性对照组OD值明显高于Bio-gide组和矿化胶原膜组,但Bio-gide组和矿化胶原膜组之间无显著性差异;7d时阴性对照组ALP活性最低,Bio-gide膜组较高,矿化胶原膜组表达最高,三者间差异有统计学意义。结论:矿化胶原膜具有良好的细胞相容性。 Objective:To investigate the biological effects of a new mineralized collagen membrane on Saos-2 cells in vitro.Methods:The Saos-2 cells were cultured on the mineralized collagen membrane and the morphology was observed by the light microscope.The extract liquid from mineralized collagen membrane and Bio-gide membrane was made.There were three groups:the blank control group,Bio-gide group and mineralized collagen membrane group.The cell proliferation and ALP activity of three groups were studied.The data were analyzed by SPSS 17.0 software.Results:Saos-2 cells on the mineralized collagen membrane readily attached to it and exhibited an elongated appearance.The MTT assay showed that the proliferation proceeded in three groups.There was no significant difference after 1,4,7 days culture among three groups in statistics.But the OD value of Saos-2 in control group were the higher than the other two groups after 10 days.The ALP activities after 7 days culture were different in the three groups.The ALP activity of mineralized collagen membrane group was the highest,and that of the blank control group was the lowest.The difference in expression of ALP among three groups was statistically significant.Conclusions:Studies in vitro revealed that the mineralized collagen membrane has good cell biocompatibility.
