探讨半枝莲醇提物对小鼠细胞色素P450酶和谷胱甘肽S-转移酶活性的影响 被引量:3

摘要 目的探讨半枝莲醇提物对小鼠细胞色素P450酶和谷胱甘肽S-转移酶活性的影响。方法于2017年2月将40只雄性SD小鼠随机分为4组,分别为对照组(10只)和试验组(30只)。试验组每天按照1 g/kg、5 g/kg、10 g/kg的用药量给所有小鼠进行灌胃,持续给药5 d。对照组予以等量生理盐水,第6天采用颈椎脱臼法处死小鼠,收集肝脏组织供后续研究,检测肝脏组织中细胞色素P450以及谷胱甘肽S-转移酶活性。结果与对照组相比,试验组低剂量组细胞色素P450含量和活性不存在差异(P>0.05),而中剂量组和高剂量组小鼠细胞色素P450含量和活性出现显著下降趋势(P<0.05)。与对照组相比,试验组小鼠谷胱甘肽S-转移酶活性没有出现显著差异(P>0.05),但中剂量组和高剂量组活性出现下降趋势。结论半枝莲醇提物对小鼠细胞色素P450酶和谷胱甘肽S-转移酶的活性有一定的抑制作用,且抑制作用随着浓度增长而不断增大,即半枝莲醇提物浓度越高,对小鼠细胞色素P450酶和谷胱甘肽S-转移酶活性抑制程度越高。 Objective To investigate the effects of ethanol extract from scutellaria barbata on cytochrome P450 and glutathione s-transferase activity in mice. Methods In February 2017, 40 male SD mice were randomly divided into 4 groups: control group(n=10) and experimental groups(n=30). Mice in the experimental group were given gavage at dose of 1 g/kg, 5 g/kg and 10 g/kg daily for 5 days. The control group was given the same amount of saline. At the sixth day,the mice were sacrificed by cervical dislocation, and the liver tissue was collected for further study. The activities of cytochrome P450 and glutathione S-transferase in the liver tissue were detected. Results Compared with the control group, there was no significant difference in cytochrome P450 content and activity in the low dose group(P〉0.05), while it showed a significant decrease in the middle and high dose group(P〉0.05). Compared with the control group, there was no significant difference in glutathione S-transferase activity in the experimental group(P〉0.05). Conclusion Ethanol extract from scutellaria barbata had certain inhibitory effect on the activity of cytochrome P450 and glutathione S-transferase in mice, and the inhibitory effect raised with the increase of concentration. It means that the higher concentration of ethanol extract from scutellaria barbata, the more inhibitory effect on activities of cytochrome P450 enzyme and glutathione S-transferase.
