摘要
[目的]对大鼠MKK3蛋白进行生物信息学分析,预测其结构和功能,可以为进一步研究大鼠MKK3蛋白的功能提供试验基础。[方法]利用生物信息学工具对大鼠MKK3蛋白的理化性质、跨膜区域、空间结构、磷酸化位点、相互作用等进行预测。[结果]大鼠MKK3蛋白由347个氨基酸残基组成,等电点为7.05,相对分子质量39.3 k D,在哺乳动物中高度保守。二级结构主要由α-螺旋、β-折叠和无规卷曲构成,结构上含一个S_TKc结构域。MKK3能与多种MAPK、Tab1、Tab2、Traf6等蛋白发生相互作用。[结论]大鼠MKK3蛋白是一种不含信号肽和跨膜区的亲水蛋白,具有激酶活性,可参与p38MAPK信号通路在炎症和肿瘤的发生发展过程中发挥重要作用。该研究为进一步研究大鼠MKK3蛋白的功能提供了试验依据。
[Objective]To predict the structure and function of rat MKK3 protein with bioinformatics.[Method]The bioinformatic tools were used to predict physical and chemical properties,transmembrane region,spatial structure,phosphorylation sites and protein-protein interaction of MKK3protein.[Result]Rat MKK3 contained 347 amino acid residues,the theoretical isoelectric point was 7.05,the calculated molecular mass was39.3k D,and it was highly conservative in mammal.The signal peptide and transmembrane regions were not found in rat MKK3.The main compositions of protein secondary structure were alpha-helix,beta-extended strand and random coil,which contained one S_TKc domain.The proteins interaction with rat MKK3 were MAPK,Tab1,Tab2,Traf6.[Conclusion]Rat MKK3 protein is a hydrophilic protein without signal peptide and transmembrane regions,the protein acts as protein kinases and involves in the activation of p38 MAPK signal pathway may play an important role in inflammation injury and tumor progress.Our research can provide useful information for the further study of rat MKK3.
