摘要
目的:观察氟比洛芬酯对切口痛大鼠疼痛过敏时脊髓丝裂原活化蛋白激酶(p38MAPK)表达的影响。方法:取成功建立切口痛模型的SD大鼠40只,随机分为5组(n=8):基础组(K组)、瑞芬太尼组(R组)、氟比洛芬酯1mg/kg组(F1组)、氟比洛芬酯2mg/kg组(F2组)及氟比洛芬酯3mg/kg组(F3组)。实验开始时K组和R组大鼠分别在腹部皮下注射生理盐水0.2ml,F1、F2、F3组大鼠分别腹部皮下注射氟比洛芬酯1mg/kg、2mg/kg、3mg/kg;5组大鼠均在戊巴比妥钠麻醉下行切口痛手术,K组大鼠切口痛手术后腹部皮下泵注生理盐水1μg/(kg·min)2h,其余4组大鼠腹部皮下泵注瑞芬太尼1μg/(kg·min)2h。所有大鼠均测定术前24h及术后6h、24h、48h左后爪的热缩足反射潜伏期(TWL);术后48h行为学检测后每组选取2只大鼠拉颈处死取L4~5脊髓处理后行Western blot检测。结果:5组大鼠切口侧跖底TWL基础值比较差异无统计学意义(P>0.05);与K组比较,R组大鼠在术后6h、24h、48h右后侧跖底TWL值降低(P<0.05)。与R组比较,F2组、F3组大鼠右后侧跖底TWL值增高(P<0.05);与K组比较,R组大鼠脊髓背角p38MAPK蛋白表达明显升高(P<0.05),与R组比较,F2、F3组大鼠脊髓背角p38MAPK蛋白表达显著降低(P<0.05)。结论:氟比洛芬酯能在一定程度上抑制由瑞芬太尼诱发的切口痛大鼠痛觉过敏时脊髓p38MAPK蛋白的表达,这可能是氟比洛芬酯减轻瑞芬太尼诱发的疼痛过敏的机制之一。
Objective : To observe the effect of flurb ip rofen axetil om expression of P38M APK : sPinal cord of incision Pain rats with hyPeralgesia induced by remifentani l. Methods: A total o f 40 SD rats w i th incision Pain were randomly d i-vided into 5 grouPs (n =8) : blank control grouP (grouP K ), remifentani l grouP (grouP R ), f lugrouP (grouP F 1), f lurbiprofen axetil 2mg/kg grouP (grouP F 2) and f lu rb ip ro fe n axetil 3mg/kg grouP (grouP F 3). A t the beginning of the exPeriment,rats in grouP K and grouP R w e re subcutaneously injected w i th normal saline 0. 2ml in abdominal skin ,group F 1,group F 2 and group F3 were subcutaneously injected w i th kg,3mg/kg respectively / groups of rats were launched incision pain surgery w i th pentobarbital sodium anesthesia,rats of group K were disposed wi th infusion of saline l %g/ ( kg * m in) 2h under abdomthe other four groups were disposed wi th infusion o f remifentani l l %g/(kg ? m in ) 2h were measured Thermal Withdrawal latency (TWL ) of the le t t paw on time 24h before operation, 6h , 24h , 48h after op-eration 2 rats in each groupwere k i l le d by p u l l in g neck in behavioral examination af ter 48h ,the L4#5 spinal c o rdw a sdetected by Western blot. Results:There was no significant difference in basal T WL value between the 5 groups P 〉 0. 05) ; compared wi th the K g ro u p, the T WL value of the r ig h t posterior plantar group ( P 〈0. 05). compared wi th the R g ro u p ,the TWL value of the r ig h t posterior plantar was increased in F2 group and F3 group ( P 〈0. 05) ; compared wi th group K , the expression of p38MA P K protein in spinal dorsal horn o f rats in R group was signif icant ly increased (P〈0. 05). Compared w i th group R ,he expression of p38dorsal horn of rats in F 2 and F3 group was sign if ica n t ly decreased (P 〈0. 05). Conclusion: F lurb ip rofe n axetil can in h ib-it the expression of p38MAPK protein in the spinal cord of incisional pain rats w i th hyperalgesia induced by remifen-tanil in a certain extent, this may be one of the mechanisms o f f lu rb ip ro fe n relieve hyperalgesia induced by remifentanil.
