父代高脂饮食对F1代雄鼠睾丸超微结构的影响

摘要 目的:探讨父代高脂饮食对F1代雄性小鼠睾丸超微结构的影响。方法:将20只3周龄C57BL/6雄性小鼠随机分为正常饮食组(CD)和高脂饮食组(HFD),分别给予普通饲料和高脂饲料。连续饲养12周后,分别与8周龄正常雌性小鼠合笼繁育F1代,在F1代15周龄大时摘取睾丸组织制作电镜切片观察超微结构。结果:CD组F1代雄鼠睾丸超微结构完整,HFD组F1代雄鼠睾丸曲细精管膜基底肿胀、厚薄不均,生精细胞层数减少、紊乱,细胞间连接疏松,胞内出现电子密度高的异常结构,线粒体出现空泡样化,精子顶体空泡。结论:高脂饮食能够对F1代雄鼠睾丸形态和生精细胞造成间接损伤,影响F1代睾丸的正常生殖功能。 Objective : To investigate the effects of hig h fa t diet on the u ltra s tru c ture of the testis of F 1 male mice.Methods: 20 C57BL /6 mice aged 3 weeks were divided into tw o groups by random numbgroup(HFD ) was fed with high fat diet while normal fa t diet g roup(CD) was fed w i th normal fatdiet. At the end of 12weeks CD/HFD F0 males were then mated wi th 8-week-old normal weight naturally cycling females to produce F1 o f--spring. At 15 weeks of age , the ultras truc ture of the testicular tissue was observed by electron microscopy. Results: T heultrastructure of the testis of F1 male mice was complete in group CD , c omparatively , which showkled basement membrane of seminiferous tubules , the layers decreased , the intercellular connections were loose , abnor-mal structures wi th high electron density appeared in the cells and vacuolization appeared in mitochondria Conclusion:High fat diet leads to indirect damage to the testis morphology and spermatogenic cells of F1 male mice , and may affectthe normal reproductive funct ion of the testis of the F1 generation.
