摘要 利用单因素实验对大肠杆菌莽草酸发酵培养基中碳氮源与金属离子进行了优化,选取对菌体生长和莽草酸产量影响较大的蛋白胨、牛肉膏、FeSO_4和MgSO_4四个因素进行正交实验优化,确定了莽草酸发酵的最适培养基配方:葡萄糖15g/L,蛋白胨10g/L,牛肉膏3g/L,酵母粉2g/L,柠檬酸2g/L,(NH_4)_2SO_41.6g/L,K_2HPO_4·3H_2O 7.5g/L,MgSO_4·7H_2O 2mg/L,FeSO_4·7H_2O 2mg/L,钼酸铵0.009mg/L,硼酸0.17mg/L,CoCl_2·6H_2O 0.047mg/L,MnSO_4·H_2O0.017mg/L,CuSO_4·7H_2O 0.017mg/L,ZnSO_4·7H_2O 0.02mg/L.采用优化后培养基发酵,莽草酸产量达到4.675g/L,为优化前的3.22倍. The carbon sources,nitrogen sources and metal ions in the medium of shikimate fermentation by Escherichia coli were optimized by single factor experiments.Four critical factors including peptone,beef extract,FeSO4·7H2O and MgSO4·7H2O were selected for further optimization using the orthogonal experiment.The optimal medium of shikimate fermentation was determined as follows:glucose 15g/L,peptone 10g/L,beef extract 3g/L,yeast powder 2g/L,citric acid 2g/L,(NH4)2SO41.6g/L,K2HPO4·3H2O 7.5g/L,MgSO4·7H2O 2mg/L,FeSO4·7H2O 2mg/L,ammonium molybdate 0.009mg/L,boric acid 0.17mg/L,CoCl2·6H2O 0.047 mg/L,MnSO4·H2O 0.017 mg/L,CuSO4·7H2O 0.017 mg/L and ZnSO4·7H2O 0.02 mg/L.Under the optimized medium,the shikimate production reached 4.675g/L,which was 3.22times as high as that obtained with the original medium.
