超临界CO2精馏结合柱层析法提纯DHA乙酯

摘要 以超临界CO_2作为流动相,结合超临界流体精馏和柱层析技术,提纯二十二碳六烯酸乙酯(DHA-EE),并考察了CO_2流速、填料颗粒大小、温度梯度以及操作压力对分离效果的影响。结果表明,当采用粒径150~250μm的硅胶填料、精馏柱从柱底到柱顶温度分布为50~75℃、CO_2流量为2.5 kg/h、萃取压力从12 MPa升至16 MPa时,在15 MPa下收集到的样品中DHA-EE的含量为92.02%,收率为10.6%。 Docosahexaenoic acid ethyl esters(DHA-EE) was purified by supercritical CO2 fractionation combined with column chromatography. The effects of CO2 mass flow rate, size of packing material, temperature gradient and pressure on the purifi cation were investigated. Under the CO2 mass fl ow rate of 2.5 kg/h, with temperature gradient from 50 to 75 ℃, pressure range from 12 to 16 MPa, the highest content of DHA-EE in samples was 92.02 % under 15 MPa, with yield of 10.6 %, using silica gel(150-250 μm mesh) as packing material.
