摘要
目的研究四氯化碳不同注射方式对构建大鼠不同分期肝纤维化模型的作用,为四氯化碳诱导大鼠不同分期肝纤维化模型提供可靠的方法。方法 42只雄性SD大鼠随机分成皮下及腹腔注射组,每周2次分别于后腿内侧皮下、腹腔注射四氯化碳-食用调和油悬浊液。第6周开始各组分别处死2~3只大鼠进行肝纤维化分期。结果皮下注射组造模期间死亡3只,死亡率14.3%;腹腔注射组造模期间死亡11只,死亡率52.4%,两种注射方式死亡率明显差异(P<0.05)。皮下注射组第7周获得有意义肝纤维化,第12周造模完成,造模完成时获得有意义肝纤维大鼠11只;腹腔注射组第6周获得有意义肝纤维化,第10周造模完成,造模完成时获得有意义肝纤维大鼠10只,两种注射方式获得有意义肝纤维化无统计学意义(P>0.05)。结论腹腔及皮下注射方式均可建立大鼠不同分期肝纤维化模型;皮下注射造模死亡率较腹腔注射低,造模完成时间较长,两种注射方式获得有意义肝纤维无明显差异。
Objective To study the effect of different injection methods of carbon tetrachloride on the establishment of different stages of hepatic fibrosis models in rats,and to provide a reliable method for carbon tetrachloride induced different stages of hepatic fibrosis models in rats.Methods 42 male SD rats were randomly divided into subcutaneous and intraperitoneal injection group,carbon tetrachloride-edible blend oil suspension was injected 2 times a week in hind leg medial skin and abdominal cavity.From the sixth week,2-3 rats were executed in each group for hepatic fibrosis staging.Results During the modeling period,3 rats died in the subcutaneous injection group,the mortality rate was 14. 3%; 11 cases died in the intraperitoneal injection group,the mortality rate was 52. 4%,and the mortality of the two injection methods was obviously different( P〈0. 05).Subcutaneous injection group used seven weeks to get significant liver fibrosis,twelve weeks to finish modeling,and obtained 11 rats with significant hepatic fibrosis.Intraperitoneal injection group used six weeks to get significant liver fibrosis,ten weeks to finish modeling,and obtained10 rats with significant hepatic fibrosis.There was no significant difference between the two injection methods obtaining significant hepatic fibrosis( P〈0. 05). Conclusion The liver fibrosis models of rats can be established by intraperitoneal and subcutaneous injection methods; the mortality of subcutaneous injection molding was lower than that of intraperitoneal injection,and the model completion time was longer.There was no significant difference between two injection methods in obtaining significant hepatic fibrosis.
