钝顶螺旋藻蛋白提取物的延缓衰老功效研究 被引量:8

Anti-aging effect of the Spirulina platensis protein extract

摘要 钝顶螺旋藻(Spirulina platensis,SP)是蓝藻的一种,蛋白含量高,广泛用于食品保健领域,目前对其活性研究主要集中在抗氧化方向,对其在延缓衰老领域的研究甚少,为了研究其延衰活性,利用秀丽隐杆线虫的优势对钝顶螺旋藻蛋白提取物(SPPE)的延衰作用进行研究。首先通过检测饲喂SPPE与对照组秀丽线虫的寿命、运动能力及脂褐素的聚集等指标来研究SPPE的延缓衰老作用,然后通过DPPH清除实验验证SPPE的体外抗氧化活性,检测SPPE对线虫在百草枯诱导的氧化胁迫下的存活时间及体内活性氧水平的影响来初步探究SPPE延衰的作用机制。结果显示,SPPE不仅能增加线虫的平均寿命和摆频,还能减少线虫体内脂褐素的聚集,并且对线虫的生殖能力没有影响。DPPH清除实验显示SPPE具有很强的体外抗氧化作用;体内实验显示,在百草枯诱导的氧化胁迫条件下,SPPE能增加线虫的存活率,降低线虫体内ROS水平。综合分析可知SPPE在秀丽线虫上具有一定的延缓衰老作用及很强的体内和体外抗氧化活性,为螺旋藻在保健食品方面开发新的功能奠定基础。 Spirulina platensis(SP) is a species of cyanobacteria that has been widely used in the field of healthy food for its high protein content. At present, most researches are focused on the SP's antioxidant activity but little on its age-delaying effect. In this study, we assess the anti-aging activities of the Spirulina platensis protein extract(SPPE) by Caenorhabditis elegans model which has advantages in anti-aging areas. We first detected the lifespan, locomotion, age pigment lipofuscin of C. elegans with or without SPPE. And then we have a preliminary study of the mechanisms of anti-aging by DPPH scavenging assay and detecting the lifespan and reactive oxygen species(ROS) of paraquat-exposed C. elegans. The results showed that SPPE could not only increase the average lifespan and the body bends of the nematodes but also reduce accumulation of the age pigment lipofuscin in the animals. No effect on the reproductive ability of the nematodes has been found. DPPH scavenging assay demonstrate the in vitro antioxidant activity of SPPE, the in vivo study showed that SPPE was able not only to increase the survival rate of the animals but also to reduce the endogenous levels of reactive oxygen species(ROS) under oxidative stress induced by paraquat. Altogether, our results demonstrate the age-delaying effect and antioxiddant activites of SPPE in vitro and in vivo, reveal a new function of Spirulina platensis on healthy food.
