仙龙颗粒对大鼠癌变模型血清中分子肝素结合性表皮生长因子、高尔基磷酸化蛋白2的影响 被引量:1

摘要 目的探讨仙龙颗粒对于血清中肝素结合性表皮生长因子(HBEGF)、高尔基磷酸化蛋白2(GOLPH2)分子表达的调控效果及仙龙颗粒的抗癌机制。 方法选择250只SD大鼠予以致癌剂构建肝癌大鼠模型,将其平均分成5组各50只,分别为阳性对照组[静脉注射顺铂(DDP),60 mg/m2]、低剂量组(喂养仙龙颗粒水溶液,1.0 g/ml)、中剂量组(喂养仙龙颗粒水溶液,2.0 g/ml)、高剂量组(喂养仙龙颗粒水溶液,6.0 g/ml)、阴性对照组(喂养蒸馏水),另取50只健康SD大鼠作空白组。对各组的大鼠进行肝脏肿瘤切除术,在手术前1周、手术后第6、12、24、36周分别按各组要求给药并每组处死1只SD大鼠作肝脏病理检查,采用酶联免疫吸附试验(ELISA)法检测剩余生存大鼠血液中的HBEGF、GOLPH2水平,统计HBEGF、GOLPH2阳性表达率。 结果死亡率方面,低剂量组术前1周为4.0%,术后36周为37.0%;中剂量组术前1周为2.0%,术后36周为2.4%;高剂量组术前1周为0.0%,术后36周为14.0%;GOLPH2阳性率,低剂量组术前1周为62.0%,术后36周为22.2%(χ^2=9.577,P=0.002);中剂量组术前1周为64.0%,术后36周为7.1%(χ^2=28.941,P=0.000);高剂量组术前1周为20.0%,术后36周为38.2%(χ^2=7.092,P=0.008);HBEGF阳性率,低剂量组术前1周为58.0%,术后36周为22.0%(χ^2=7.666,P=0.006);中剂量组术前1周为62.0%,术后36周为16.7%(χ^2=17.525,P=0.000);高剂量组术前1周为56.0%,术后36周为29.4%(χ^2=4.752,P=0.029)。不同剂量的仙龙颗粒均能显著降低大鼠的死亡率、血清中HBEGF、GOLPH2的阳性率,但低剂量组、高剂量组的效果不及中剂量组。 结论仙龙颗粒可下调肝癌大鼠血清中HBEGF、GOLPH2的表达,从而抑制癌细胞增殖及促进其凋亡,达到抗癌、延长存活时间的效果。 ObjectiveTo explore Xian Long particles for heparin-binding epidermal growth factor (HBEGF), golgi phosphorylated protein 2 (GOLPH2) protein regulation effects and anticancer mechanism fairy dragon particles. Methods250 cases of SD mice with induced liver cancer were selected as the test group, and 50 cases of healthy mice were selected as the blank group, and different doses of Xian Long particles were fed, the mortality rates, positive rates of HBEGF and GOLPH2 after tumorectomy were compared. ResultsThe mortality rate was 4.0% in one week before intervention, 37.0% in 36 weeks after intervention (χ2=9.577, P=0.002), 2.0% in one week before intervention and 2.4% in 36 weeks after intervention (χ2=28.941, P=0.000). Week after operation. The positive rate of GOLPH2 was 62.0% in one week and 22.2% in 36 weeks after operation (χ2=7.092, P=0.008). The GOLPH2 positive rate was 64.0% in one week after operation The positive rate of HBEGF was 58.0% in one week before intervention and 22.0% in 36 weeks after intervention (χ2=7.666, P=0.006), and the intervention group 62.0% at 1 week before operation and 16.7% at 36 weeks after operation (χ2=17.525, P=0.000), and 56.0% at 1 week before operation and 29.4% at 36 weeks after operation (χ2=4.752, P=0.029). Different doses of fairy dragon particles can significantly reduce mortality in rats, serum HBEGF, the positive rate of GOLPH2, but the effect of low-dose group, high dose group of less than middle dose group. ConclusionXian Long particles may cut liver cancer in rats of HBEGF, GOLPH2 protein.
