摘要
目的探讨去卵巢老年大鼠雌激素下降与软骨退变的相关性。方法 24只6月龄雌性SD大鼠按数字表法随机分为对照组、去卵巢组和去卵巢+跑步组各8只。对照组常规饲养,去卵巢组行双侧卵巢切除术,去卵巢+跑步组行双侧卵巢切除术合并疲劳训练。5周后处死,取左侧膝关节石蜡包埋,行HE染色、免疫组化分析、膝关节软骨Mankin评分,右侧膝关节抽取关节液检测TNF-α含量,取血清检测E_2含量。结果 (1)膝关节切片HE染色显示:与对照组比较,去卵巢组与去卵巢+跑步组膝关节软骨均出现缺损,其中去卵巢+跑步组缺损最重;(2)与对照组比较,去卵巢组和去卵巢+跑步组mankin评分、膝关节MMP-13表达、关节液TNF-α浓度均明显提高(P<0.01),血清E_2浓度明显降低(P<0.01),且以去卵巢+跑步组变化最为显著。结论老年大鼠雌激素的下降与软骨退变具有一定的相关性。
Objective: To investigate the relationship between estrogen decline and cartilage degeneration among ovariectomized aged rats. Methods: Twenty-four female 6-month-old SD rats were randomly divided into control group, ovariectomized group and ovariectomized + running group according to random number table. The control group was fed with conventional diet, the ovariectomized group was treated with bilateral ovariectomy, and the ovariectomized+running group received bilateral ovariectomy combined with fatigue training. Rats were executed after 5 weeks, and their left knees were treated with paraffin-embedded, HE staining, immunohistochemical analysis, knee joint cartilage mankin score. The synovial fluid of right knee joint extract was extracted to detect the TNF-α, and E2 was determined through the serum. Results: (1) HE staining of knee joint section showed that there were defects in articular cartilage in ovariectomized group and ovariectomized +running group, compared with the control group, especially in ovariectomized running group. (2) Compared with the control group, Mankin score, expression of MMP-13 in knee joint and the concentration of TNF-α of synovial fluid in ovariectomized group and ovariectomized+ running group were significantly increased (P〈 0.01, P〈0.05). Serum E2 concentration was significantly decreased (P〈0.01, P〈0.05). Conclusion: The decline of estrogen in old rats is related to the degeneration of cartilage.
