血必净对重症急性胰腺炎患者氧化应激的影响

Effects of Xuebijing on oxidative stress in patients with severe acute pancreatitis

[目的]探讨血必净对重症急性胰腺炎患者氧化应激的影响。[方法]取2016年1月~2016年12月我院重症医学科收治的16位SAP患者的血清,将原代人脐静脉内皮细胞(HUVEC)传代培养后接种于96孔培养板,分为空白对照组、血清处理组和血必净治疗组(1mg/ml、5mg/ml、25mg/ml)进行培养,然后分别测定各组的氧化应激指标ROS、MDA、SOD和T-AOC。[结果]ROS、MDA在血清处理组明显高于空白对照组[ROS:(97.22±6.45)U/ml(53.08±5.28)U/ml,(32.75±1.24)μmol/L(12.05±1.48)μmol/L,P<0.05],血必净治疗组ROS、MDA水平随着血必净浓度增加逐渐降低,与血清处理组存在显著差异。SAP患者血清处理后的SOD、TAOC水平均低于空白对照组[(25.54±1.17)U/ml(39.56±1.13)U/ml,(2.12±0.09)U/ml(3.22±0.05)U/ml,P<0.05],而给予不同血必净治疗后SOD和T-AOC较血清处理组增高。[结论]血必净可以有效控制SAP患者的氧化应激反应,减轻炎症反应,可用于SAP患者的治疗。

[Objective]To investigate the effect of Xuebijing on oxidative stress in patients with severe acute pancreatitis（SAP）.[Methods]The serum specimens of 16 SAP patients in ICU of the first hospital of Lanzhou university were preserved.The primary human umbilical vein endothelial cells（HUVEC）were subcultured and inoculated in 96-well culture plates.All the cells were divided into the control group,the serum group and the Xuebijing treatment group（1 mg/ml,5 mg/ml,25 mg/ml）,and then the parameters of oxidative stress were measured by ROS,MDA,SOD and T-AOC.[Results]ROS and MDA in serum group was significantly higher than those of the control group[ROS：（97.22±6.45）U/ml vs.（53.08±5.28）U/ml,（32.75±1.24）μmol/L vs.（12.05±1.48）μmol/L,P〈0.05].The levels of ROS and MDA in the Xuebijing treatment group decreased gradually as the concentration of Xuebijing increased,and there was a significant difference with the serum group.The levels of SOD and T-AOC in the serum group were lower than those of the control group[（25.54±1.17）U/ml vs.（39.56±1.13）U/ml,（2.12±0.09）U/ml vs.（3.22±0.05）U/ml,P〈0.05）].After treatment with different concentration of Xuebijing,SOD and TAOC were higher than those of serum group.[Conclusion]Xuebijing can effectively control the oxidative stress and alleviate the inflammatory reaction in patients with SAP,and can be used in the treatment of SAP patients.
