摘要
为探究中药复方多糖对不同MHC B-LβⅡ基因型鸡的免疫调节作用,采用PCR-SSCP方法将黄麻肉鸡按不同MHC B-LβⅡ基因型分组后于28日龄时各组随机取5只试验鸡,采集外周血,分离淋巴细胞,与终浓度为0.5、1、2μg/mL的TLR2抗体共培养1h后,分别加入终浓度为200、100、50、25、0μg/mL的中药复方多糖共培养24h,采用ELISA方法检测不同MHC B-LβⅡ基因型鸡外周血淋巴细胞培养上清液中IL-6、TNF-α和NF-κB的质量浓度。结果显示,加入TLR2抗体后,中药复方多糖促进淋巴细胞分泌IL-6、TNF-α和NF-κB的能力受到明显抑制,并且随着TLR2抗体浓度的增加,抑制作用增强。表明TLR2是中药复方多糖的免疫调节作用受体之一,中药复方多糖可能通过与淋巴细胞表面TLR2结合,经TLR2/NF-κB信号通路活化转录因子NF-κB,促进IL-6和TNF-α的分泌发挥其免疫调节作用。
In order to investigate the immunomodulatory effects of traditional Chinese medicine compound polysaccharides(cCHMPS) in chickens with different MHC B-LβⅡ genotypes,PCR-SSCP technique was applied to group HuangMa chickens according to different MHC B-LβⅡ genotypes. On 28 days, five chickens' blood for each group were collected and the lymphocytes were isolated and cultured with final concentrations of 0.5 μg/mL, 1 μg/mL,2 μg/mL of TLR2 antibody for 1 h,then cCHMPS were added to the final concentrations of 200 μg/mL, 100 μg/mL, 50 μg/mL, 25 μg/mL, 0 μg/mL, respectively and co-cultured for 24 h.ELISA method was used to detect the concentrations of IL-6 ,TNF-α and NF-κB in lymphocyte culture supernatants.The results showed that cCHMPS could promote the secretions of IL-6 ,TNF -α and NF-κB in lymphocytes of chickens with different MHC B-Lβ Ⅱ genotypes, cCHMPS could inhibit the secretions of IL-6 ,TNF-α and NF-κB in lymphocytes when co-cultured with TLR2 antibody,and with the increase of the concentrations of antibody,the inhibition increased.It suggested that TLR2 is one of the immunomodulatory receptors of cCHMPS, cCHMPS is combined with the cell surface TLR2 receptor, activates the transcription factor NF-κB by TLRs/NF-κB signaling pathway, promotes the secretions of cytokines IL-6, TNF-α to regulate immune effects.
