摘要
目的研究N25对肺癌H460、H1299、A549细胞增殖、细胞周期、基因表达的影响,并探讨其抗肿瘤的作用机制。方法 CCK-8法检测不同浓度的N25作用H460、H1299、A549细胞48h的增殖作用;流式细胞术检测N25对H460的细胞周期阻滞情况;RT-PCR检测N25作用H460细胞48h后,对p21W A F1/CIP1、p53基因表达的影响。结果 CCK-8法表明N25对H460、H1299、A549细胞生长的抑制作用呈现剂量依赖性;流式细胞术结果显示,N25阻滞H460细胞周期于G2/M期;RT-PCR检测表明,它能明显下调H460细胞的p21W A F1/CIP1、p53的基因表达;结论 N25可抑制H460、H1299、A549细胞的增殖,并使得H460细胞株的细胞周期阻滞于G2/M期。其抗肿瘤的机制可能与下调P21W A F1/CIP1、P53表达相关。
OBJECTIVE To investigate the effects of compound N25 on cell proliferation, cell cycle, gene expression in lung cancer H460, H1299 ,A549 cells and to explore its anti-tumor relative mechanisms. METHODS CCK-8 assay was used to investigate the cell proliferation by different concentrations of N25 treated with lung cancer cell for 48 hours. Flow cytometry was performed to detecte N25's effect on the cell cycle of H460 cell. Furthermore, RT-PCR method was used to evaluate gene expression of p21WArl/CIP1 and p53 in H460 cell with N25 incubated for 48 hours. RESULTS N25 demonstrated a dose dependent manner of growth inhibition in three lung cancer cells lines. Flow cytometry results showed that N25 could arrest the cell cycle in G2/M phase in H460 cell. RT-PCR analy- sis indicated N25 significantly down-regulated ihe gene expression of p21wArl/cIPl and p53. CONCLUSION N25 could inhibit the proliferation of lung cancer H460, H1299, A549 cells and arrested cell cycle in G2/M phase in H460 cell. Its anti-tumor mechanism might be relate to down-regulate the expression of p21wArl/cIP1 and p53.
