摘要
以平贝母为实验材料,考察并优化平贝母总生物碱的提取工艺。以贝母素乙为对照品,采用酸性染料比色法在412nm波长处测定平贝母中总生物碱的含量,比较加热回流、超声及冷浸提取法对总生物碱的提取率,选取最佳提取工艺。结果表明:NH_3·H_2O 3mL,浸润2h,50℃恒温超声提取25min,再超声45min,提取效率最佳,生物碱得率为0.2347%。即超声与加热回流相结合的提取工艺稳定可行且提取效率较高,可作为平贝母总生物碱的提取工艺。
Bulbus Fritillariae was taken as test materials,the extraction process of total alkaloids from Fritillaria ussuriensis Maxim.was studied and optimized.Taking the yield of total alkaloids as control sample, the content of total alkaloids in Fritillaria was determined at 412nm wavelength by acid dye colorimetry.Selecting the best extraction process by comparing heating reflux,ultrasonic and cold-maceration extraction.The results show that the optimum extraction rate is that the ammonia water is 3mL and the infiltration time is 2h,under the constant temperature of 50℃, the ultrasonic extraction of 25min, and then ultrasound 45min, the alkaloid yield is 0.2347%.The extraction process,which combines ultrasonic and reheating reflux, is stable and efficient,and can be used as the extraction process of the total alkaloid of Fritillaria.
