安宫牛黄丸对脑出血后神经功能与肠粘膜屏障的影响 被引量:14

Effect of Angong Niuhuang pill on intestinal mucosal barrier in intracerebral hemorrhage mice

摘要 目的:研究安宫牛黄丸对脑出血后神经功能与肠粘膜屏障通透性的影响。方法:将50只雄性CD-1小鼠随机分为假手术组、模型组(脑出血)、安宫牛黄丸0.1、0.2和0.4 g/kg组。脑立体定位仪注射胶原酶制备小鼠脑出血模型;Garcia test评价小鼠神经功能;干湿重法检测脑组织含水量;FITC-D法检测小鼠肠粘膜通透性;HE染色法观察肠道病理学改变;ELISA法检测血浆内毒素含量。结果:安宫牛黄丸0.2、0.4g/kg可改善脑出血小鼠神经功能,能够降低肠粘膜通透性、减轻肠粘膜损伤;安宫牛黄丸0.1、0.2、0.4 g/kg显著减轻脑水肿,降低血浆内毒素含量。结论:安宫牛黄丸可改善脑出血后的神经功能;减轻脑出血所致的肠粘膜损伤,降低肠粘膜通透性,这可能是其治疗脑出血的作用机制之一。 Objective: To investigate the effect of Angong Niuhuang pill on neurological function and intestinal mucosal barrier in intracerebral hemorrhage mice. Methods: Fifty male CD-1 mice were randomly divided into the sham group,intracerebral hemorrhage group,0. 1 g/kg Angong Niuhuang pill group,0. 2 g/kg Angong Niuhuang pill group and 0. 4 g/kg Angong Niuhuang pill group. The intracerebral hemorrhage mice were prepared by stereotaxic injection of collasenase. Garcia test was used to evaluate the neurological function of mice. The water content of brain tissue was measured by dry and wet weight method. Permeability of intestinal mucosa was detected by FITC-D method. The pathological changes of intestine were also observed. The ELISA kit was used to detect the plasma endotoxin. Results: Angong Niuhuang pill(0. 2,0.4 g/kg) improved neurological function,decreased intestinal permeability,and attenuated intestinal mucosal injury in intracerebral hemorrhage mice. It also showed that Angong Niuhuang pill(0. 1,0. 2,0. 4 g/kg) reduced brain edema and plasma endotoxin. Conclusion: Angong Niuhuang pill can ameliorate the neurological function and protect the intestinal mucosal barrier,reduce the absorption of endogenous endotoxin,which may be one of the mechanisms of its treating patients with intracerebral hemorrhage.
