摘要
目的:研究β-榄香烯对不同分子亚型人乳腺癌细胞增殖、侵袭和迁移的作用。方法:以不同转移潜能MCF-7、MDAMB-231乳腺癌细胞作为研究对象,设空白对照组及药物组,药物组以不同浓度β-榄香烯溶液作用细胞24h与48h后,CCK8法检测其对细胞活力的影响。应用细胞克隆形成实验进一步检测β-榄香烯对两种细胞增殖和克隆形成能力的影响。此外,Transwell实验和划痕实验检测β-榄香烯对高转移性细胞株MDA-MB-231细胞侵袭和迁移能力的作用。结果:β-榄香烯对MCF-7、MDA-MB-231乳腺癌细胞的活力均有抑制作用,且对高转移细胞MDA-MB-231抑制作用更强,对于MCF-7细胞24h,48h的半数抑制浓度(IC50)分别为(43.45±0.43),(43.05±0.56)μg/ml,对MDA-MB-231细胞24h,48h的半数抑制浓度(IC50)分别为(12.20±0.61),(8.30±0.27)μg/ml。克隆形成实验结果表明β-榄香烯能够抑制乳腺癌细胞的克隆形成率,与空白对照组相比具有显著性差异。当β-榄香烯浓度为25μg/ml时,对MDA-MB-231细胞的抑制率达到49.2%,对MCF-7只有15.5%。此外,Transwell实验和划痕实验结果显示当β-榄香烯浓度达到10μg/ml时就能抑制高转移性细胞株MDA-MB-231的侵袭和迁移能力。结论:β-榄香烯能够抑制MCF-7、MDA-MB-231的细胞活力、增殖和克隆形成,且对高转移性细胞株MDA-MB-231的侵袭和迁移能力显示出很强的抑制作用,其具体作用机制有待进一步研究。
Objective: To investigate effects of β-elemene on proliferation,invasion and migration of human breast cancer cells with different molecular subtypes. Methods: MCF-7 and MDA-MB-231 with different metastatic potential were used as research objects. After cancer cells were incubated with different concentrations of β-elemene solution for 24 h or 48 h,the cell viability was detected by CCK8. Effects of β-elemene on the proliferation and colony forming ability of two kinds of cells were further examined by cell cloning assay. In addition,Transwell assay and scratch assay were used to detect the effect of β-elemene on invasion and migration ability of MDA-MB-231 cell line. Results: β-elemene inhibited cell viabilities of MCF-7 and MDA-MB-231,and it had the stronger inhibition on high metastatic MDA-MB-231. Half inhibitory concentrations(IC50) of MCF-7 at 24 h and 48 h were(43. 45 ± 0. 43) and(43. 05 ± 0. 56) μg/ml respectively,IC50 of MDA-MB-231 were(12. 20 ± 0. 61) and(8. 30 ± 0. 27) μg/ml respectively. The result of cloning experiment showed that β-elemene significantly inhibited the clone formation rate of breast cancer cells,compared with the control group(P〈0. 01). When the concentration of β-elemene was 25μg/ml,the inhibitory rate of MDA-MB-231 cells reached 50%,only 15. 5% for MCF-7. In addition,Transwell test and scratch test showed that it inhibited the migration and invasion of the highly metastatic cell line MDA-MB-231 when β-elemene concentration reached 10μg/ml,and compared with the control group,there was the significant difference(P〈0. 01). Conclusion: β-elemene can inhibit cell viabilities,proliferations and colony formations of MCF-7 and MDA-MB-231,and shows a strong inhibitory effect on invasion and migration of the high metastatic MDA-MB-231,but the specific mechanism needs further to be studied.
